基于核酸适配体的胶体金分光光度法检测赭曲霉毒素A 被引量:7

Spectrophotometric detection of ochratoxin A based on aptamer and gold nanoparticles

摘要 基于核酸适配体对靶标的特异性和胶体金分光光度法的灵敏性,建立了检测赭曲霉毒素A(OTA)的方法。利用核酸适配体能够抑制胶体金在高盐溶液中的聚集,当OTA存在时,其与适配体特异性结合,使胶体金发生聚集,胶体金的颜色发生变化。对NaCl浓度、核酸适配体浓度、反应时间等条件进行了优化。在最优的条件下,吸光度的比值(A6 56 nm/A52 0 nm)与OTA质量浓度在0~300 ng/mL的范围内呈良好的线性关系,其线性回归方程为y=0. 001x+0. 2264,相关系数R2为0. 9962,加标回收率为99. 2%~103. 1%,相对标准偏差为4. 4%。方法可作为OTA的快速检测方法。 A simple and rapid method for the detection of ochratoxinA( OTA) was established based on the specificity of the aptamer for the target and the sensitivity of gold nanoparticles( AuNPs) by spectrophotometry. The aptamer can inhibit the aggregation of Au NPs in high salt solution. In the presence of OTA,it binds specifically with the aptamer,causing the Au NPs to aggregate and the color to change. In this experiment,salt concentrations,aptamer concentrations,the reaction time and other conditions were optimized. Under optimal conditions,the mass concentration of OTA in the range of0-300 ng/m L exhibited a good linear relationship with the ratio of absorbance( A656 nm/A520 nm). The linear regression equation was y = 0. 001 x + 0. 2264,the coefficient R2 was 0. 9962,the spiked recoveries were 99. 2%-103. 1%,and the relative standard deviation was 4. 4%.
