摘要
目的观察不同时间穴位贴敷对过敏性哮喘大鼠行为学及血清免疫球蛋白E(IgE)、白细胞介素-4(IL-4)和干扰素-?(IFN-?)水平的影响,探讨穴位贴敷治疗哮喘的作用机制。方法 50只Wistar雄性大鼠按体重采用随机数字表法分为A组(正常对照组)、B组(哮喘模型组)、C组(地塞米松组)、D组(穴位贴敷2 h组)和E组(穴位贴敷4 h组)。以鸡卵白蛋白(OVA)制备大鼠哮喘模型,14 d后各组分别给予相应治疗。观察大鼠治疗前后引喘潜伏期和发作总持续时间变化;采用HE染色检测大鼠肺组织病理改变,ELISA法测血清中IgE、IL-4和IFN-?水平变化。结果与A组比较,B组引喘潜伏期显著缩短、发作持续时间显著延长(P<0.05);病理显示肺组织炎症明显;IgE、IL-4显著升高,IFN-?显著降低(P<0.05)。与B组比较,C组、D组、E组治疗后引喘潜伏期明显延长、哮喘发作持续时间明显缩短(P<0.05);肺病理组织观察显示穴位贴敷后支气管及肺实质炎性改变显著改善;血清中IgE和IL-4水平显著降低,IFN-?水平显著升高(P<0.05)。D组、E组各指标比较,差异均无统计学意义(P>0.05)。结论穴位贴敷可以通过改善肺组织炎症,减低Ig E、IL-4,升高IFN-?的表达来治疗哮喘,且不同贴敷时间对治疗效果无明显影响。
Objective To observe the effect of acupoint application at different time points on the behaviors and levels of immunoglobulin E (IgE), interleukin (IL)-4 and interferon (IFN)-γ in allergic asthma rats, and to discuss the mechanism of acupoint application in treating asthma. Method Based on the body weight, fifty male Wistar rats were divided by the random number table into group A (normal control group), group B (asthma model group), group C (dexamethasone group), group D (acupoint application 2 h group) and group E (acupoint application 4 h group). Asthma rat model was developed by ovalbumin (OVA), and each group received corresponding intervention 14 d later. The attack latency and duration were observed before and after the intervention;HE staining was used to detect the pathological changes in lung tissues;enzyme-linked immunosorbent assay (ELISA) was used to examine the changes in serum levels of IgE, IL-4 and IFN-γ. Result Compared with group A, the asthma attack latency was significantly shortened and the attack duration was significantly extended in group B (P<0.05);pathological analysis showed obvious pulmonary inflammation;the levels of IgE and IL-4 increased markedly and the level of IFN-γ decreased significantly (P<0.05). Compared with group B, the attack latency was significantly extended and the duration was markedly shortened in group C, D and E (P<0.05);pathological analysis revealed improved bronchial and pulmonary inflammation;the serum levels of IgE and IL-4 decreased significantly and the level of IFN-γ increased significantly (P<0.05). There were no significant differences in the indexes between group D and E (P>0.05). Conclusion Acupoint application can treat asthma through improving pulmonary inflammation, down-regulating IgE and IL-4 and up-regulating IFN-γ, and different application time does not have significant impact on the treatment result.
