摘要
[目的]通过建立高效液相色谱法(HPLC)进行棉籽中游离棉酚的含量测定。[方法]样品用乙醇提取,用HPLC进行检测。液相条件为:Agilent SB-C18(250 mm×4.6 mm,5μm);流动相为甲醇∶0.1%磷酸=85∶15,检测波长235 nm,柱温35℃,流速0.8 m L/min,进样量10μL。[结果]游离棉酚在1.06~42.40μg/m L线性关系良好,精密度、稳定性、重复性的RSD<2%。对全疆20个棉籽进行含量测定,样品含量为25~4 887 mg/kg。[结论]该方法具有良好的稳定性、准确度、灵敏度和回收率,可以适用于棉籽中游离棉酚的含量测定。
[Objective]The determination method of free gossypol(FG)in cotton-seed meal was established by high perform ance liquid chrom atography(HPLC).[Method]The cotton-seed samples were extracted by ethanol,and detected by HPLC.The chromatographic conditions were as follows:the column was Agilent SB-C 18(250 mm×4.6 mm,5μm).The mobile phase was methanol-0.1%H 3PO 4(85∶15),and the UV detection wavelength was 235 nm,the column temperature was 35℃,the flow rate was 0.8 mL/min,the sample volume was 10μL.[Result]The free gossypol has a good linear relationship at 1.06~42.40μg/mL.Precision,stability,repeatability and recovery experiments met the requirements for the determination and RSD<2%.The content of 20 cotton-seed in Xinjiang was determined,and the sample content was 25-4 887mg/kg.[Conclusion]The method has good stability,accuracy,sensitivity and recovery,and it can be applied to the determination of free gossypol in cotton-seed.
