摘要
以湖南本地种的蕙兰种子和春兰新芽为外植体,以75%乙醇、0.1%升汞、10%次氯酸钠为消毒剂设计了6个消毒方案进行外植体消毒试验。结果表明:蕙兰种子组织培养时采用75%酒精45 s+0.1%升汞消毒8 min的效果较佳,75%酒精45 s+0.1%升汞消毒5 min对春兰新芽组培灭菌效果较好;同时,升汞的消毒效果总体优于次氯酸钠,但需要掌握消毒时间,以免过度处理给外植体造成伤害导致其死亡。
Taking Hunan local orchid seeds and buds as explants,the explants disinfection tests were conducted by use 75%ethanol,0.1%mercuric chloride,10%sodium hypochlorite as disinfectant recpectively..The results showed that the organization of Cymbidium seed culture by using 75%alcohol 45 s+0.1%mercuric chloride for 8 min was the best,75%alcohol 45 s+0.1%mercuric chloride for 5 min ideal sterilization effect on tissue culture of Cymbidium shoots.The disinfection effect of mercuric chloride was better than that of sodium hypochlorite,but need to master the disinfection time,so as to avoid excessive damage to the explants.
