电针预处理对脂多糖诱导的急性肺损伤大鼠炎性细胞因子及水通道蛋白5的影响 被引量:5

Effect of Electroacupuncture Pretreatment on Inflammatory Cytokines and Aquaporin-5 in Acute Lung Injury Rats Induced by Lipopolysaccharide

摘要 目的探讨电针足三里穴预处理对脂多糖(LPS)诱导的急性肺损伤(ALI)大鼠的干预作用及可能机制。方法将SD大鼠随机分为正常、ALI4h、ALI6h、针刺+ALI4h和针刺+ALI6h组,ALI4h、ALI6h组大鼠气管内用LPS(2mg/kg)分别滴注4h和6h以诱导ALI,后两组大鼠在针刺预处理双侧足三里穴1周后于气管内分别滴注LPS4h和6h。各组均取血、支气管肺泡灌洗液(BALF)及肺组织,计算BALF中白细胞总数及肺湿/干比(W/D),HE染色观察肺组织形态,ELISA方法检测血浆中AQP_5、IL-1β、IL-6、IL-10、TNF-α和BALF中AQP_5水平,免疫组化方法观察肺组织AQP_5表达。结果与正常组比较,ALI4h组白细胞计数、W/D增高(P<0.05)、肺泡总面积和肺组织面积之比(A/t)降低(P<0.05)以及IL-1β、IL-10、TNF-α含量显著增高(P<0.001);ALI6h组IL-6和IL-10含量明显增高(P<0.01);ALI4h、ALI6h组AQP_5含量显著降低(P<0.001)、积分光密度(IOD)明显降低(P<0.01)。与ALI4h组比较,针刺+ALI4h组白细胞计数与W/D降低(P<0.05)、IL-1β与TNF-α含量降低(P<0.05)、血浆及BALF中AQP_5含量均升高(P<0.05),针刺+ALI6h组血浆AQP_5含量有升高(P<0.05)。结论电针足三里穴预处理对LPS诱导的ALI大鼠具有抗炎、降低肺水肿的保护作用,且在诱导4h时的ALI中更佳,其机制可能与电针下调炎性细胞因子和上调AQP_5的表达有关。 Objective To investigate the influence of the pretreatment of electroacupuncture(EA)on the inflammatory cytokinesand aquaporin(AQP)-5in acute lung Injury rats induced by lipopolysaccharide.Methods SD-rats were randomly divided into5groups:control,ALI4h,ALI6h,EA plusALI4h and EA plus ALI6h.Acute lung injury was induced by LPS(2mg/kg)stilling into the trachearespectively for4h and6h except the normal group.The rats in latter two groups were pretreated with EA at bilateral Zusanli(ST36)points for a week.The plasma was separated for detecting the content of AQP5,IL-1茁,IL-6,IL-10and TNF-琢by enzyme-linkedimmuno sorbent assay(ELISA)method.Bronchoalveolar lavage fluid(BALF)of rats was extracted for the content of AQP5by ELISA andcounting the total number of white blood cells(WBC).The left lung tissue was used for the ratio of wet to dry weight(W/D),andright lung was fixed in formaldehyde for observation of morphology and immunohistochemistry.Results Compared with the control group,in ALI4h group W/D and WBC count was increased(P<0.05),the ratio of total area of alveolar to lung tissue area(A/t)was decreased(P<0.05),IL-10,TNF-Band IL-1awere significantly increased(P<0.001);and IL-6and IL-10were significantly higher(P<0.01)in ALI6h group;the content of AQP5decreased significantly(P<0.001),integral optical density(IOD)decreased significantly(P<0.01)in ALI4h and ALI6h groups.Compared with ALI4h group,WBC count and W/D decreased(P<0.05),IL-1Band TNF-acontent decreased(P<0.05),AQP5content in plasma and BALF increased(P<0.05),and plasma AQP5content in acupuncture group was increased in EA plus ALI4h group.Conclusion EA pretreatment at Zusanli could protect ALI rats induced LPS from inflammatory and reduce pulmonary edema,and the effect was better in ALI4h group than in ALI6h group.The mechanism may be related to the reduction of inflammatory cytokines and the up-regulation of AQP5expression by EA pretreatment.
