短期针刺对急性肺损伤大鼠血浆白细胞介素-1β含量及海马c—fos基因表达的影响 被引量:10

Effect of short-term acupuncture on content of plasma interleukin-1β and expression of c-fos proto-oncogene in hippocampus of rats after acute lung injury

摘要 目的探讨针刺对急性肺损伤(ALI)的干预作用及其可能机制。方法将36只Wistar大鼠按随机数字表法分为对照组、模型组、假针刺组、针刺组,每组9只。采用经尾静脉注射油酸0.2ml/kg后4h注射脂多糖(LPS)2mg/kg的“二次打击”复制大鼠ALI模型。针刺组于制模前5d每日针刺大鼠肺俞、足三里穴;假针刺组针刺离穴位5mm非经非穴处;对照组和模型组不给予任何治疗。于制模后2h取血检测大鼠动脉血氧分压(PaOz)及血浆白细胞介素-1β(IL-1β)含量;取肺组织进行病理观察及形态学积分测定;取脑组织检测海马CA2-3区c-fos表达。结果与对照组比较,模型组Pa02[mmHg(1mmHg=0.133kPa):57.56±5.61比107.22±3.233明显降低(P〈0.01),血浆IL-1β含量(μg/L:0.76±0.06比0.25±0.02)、肺组织病理形态学积分(分:12.16±0.82比1.68±0.14)及海马c-fos阳性表达(灰度值:94.22±7.89比189.28±8.45)均明显升高(均P〈0.01)。针刺组PaO2[(64.56±5.77)mmHg]改善及血浆IL-1β含量[(0.71±0.05)μg/L]下降,肺损伤减轻[病理形态学积分(11.58±0.50)分]及海马c-fos阳性表达(灰度值129.16±8.73)明显减少,与模型组比较差异有统计学意义(P〈O.05或P〈0.01)。结论大鼠海马CA2~3区通过c-fos途径参与ALI免疫调节过程;针刺对ALI具有一定的保护作用,其机制可能与降低血浆IL-1β含量、抑制c-fos表达有关。 Objective To explore the interventional action and possible pathogenesis of acupuncture in acute lung injury (ALl). Methods Thirty-six Wistar rats were divided into four groups: control, model, sham acupuncture and acupuncture groups by means of random numeral table, with 9 rats in each group. A "two hit" animal model of ALI in rats was induced with injection of 0.2 ml/kg oleic acid and 4 hours later with the injection of 2 mg/kg lipopolysaccharide (LPS) via a caudal vein. Manual acupuncture on Feishu point (肺俞穴) and Zusanli point (足三时穴) was performed in rats of acupuncture group before modeling 5 days. The needle stimulation in sham acupuncture group was made by puncturing points (non-meridian-non-acupoint 3 approximately 5 mm lateral to Feishu point or Zusanli point. No treatment was given in control gorup or model group. Two hours later after modeling, the rats' blood samples were obtained to detect arterial partial pressure of oxygen (PaO2) and content of plasma interleukin-1β (IL-1β); lung tissue samples were collected to examine their pathological changes and count morphological scores; cerebral tissue samples were taken to measure the c-fos expression in the CA 2 - 3 region of hippocampus in rats. Results Compared to the control group, the PaO2(mm Hg, 1 mm Hg=0.133 kPa, 57.56i5.61 vs. 107.22±3.23) was decreased markedly (P〈0.01), while the plasma IL-1β content (μg/L: 0. 76±0.06 vs. 0.25±0.02), pathological morphological score (12.16±0.82 vs. 1.68± 0. 14) and hippocampus c-fos positive neurons expression (the gray values: 94.22±7.89 vs. 189.28±8.45) were increased distinctly in the model group (all P〈0. 01). The PaO2 ((64.56± 5.77) mm Hg] and plasma IL-1β content 〈(0. 71±0. 05) μg/L] were improved, the lung injury was also alleviated (pathological morphological score: 11.58± 0.50), the hippocampus c-fos expression was decreased significantly in the acupuncture group (gray value: 129.16±8.73), and the differences were significant in comparison with those of the model group (P 〈 0.05 or P 〈 0. 01). Conclusion Via c-los proto-oncogene pathway, the CA2- 3 region of hippocampus in rats may participate in the immune regulatory process of ALI; acupuncture can exert protective effects on ALl, and its mechanism might be related to the decrease of the content of plasma IL-1β and the inhibition of the expression of c-fos gene in the CA2 - 3 region of hippocampus.
