过氧化物酶1在卵巢上皮性浆液性肿瘤中的表达及其临床意义 被引量:4

Expression of PRDX1 in epithelial ovarian serous tumor and its clinical significance

摘要 目的:检测过氧化物酶1(peroxiredoxin 1,PRDX1)在卵巢上皮性浆液性癌中的表达,探讨并预测PRDX1参与肿瘤发生发展的机制及其临床意义。方法:采用免疫组化SP法检测55例卵巢上皮性浆液性乳头状癌、11例卵巢上皮性交界性肿瘤、21例卵巢上皮性良性肿瘤以及15例正常卵巢组织中PRDX1的表达情况,利用单因素和多因素Cox回归模型分析其与卵巢上皮性浆液性乳头状癌临床病理参数及预后的关系。从癌症基因组图谱(TCGA)数据库收集374个卵巢癌样本,并对PRDX1进行基因富集分析(gene set enrichment analysis,GSEA)。结果:PRDX1以胞浆着色为主。PRDX1在卵巢上皮性浆液性乳头状癌中的阳性率(96.36%)明显高于卵巢上皮性交界性肿瘤(72.72%)、卵巢上皮性良性肿瘤(57.14%)及正常卵巢组织(20%)(P均<0.05)。在卵巢上皮性浆液性乳头状癌中,PRDX1表达随FIGO分期增加而增加(P<0.05),PRDX1为影响卵巢上皮性浆液性癌预后的独立危险因素(P<0.05)。基因富集分析发现PRDX1高表达样本富集到氧化磷酸化基因集,PRDX1低表达样本富集到Wnt信号通路、ErbB信号通路、Notch信号通路、癌症信号通路、黏附桥接等基因集。结论:PRDX1参与调控肿瘤Wnt、ErbB、细胞黏附、氧化应激等信号通路,其表达与卵巢癌的发生、发展及预后相关,可作为卵巢癌预后的标记物。 Objective:To identify the expression of PRDX1 (peroxiredoxin 1) in epithelial ovarian serous carcinoma(EOSC),and to analysis its correlations with the clinical features and prognosis of ovarian cancer.Methods:Immunohistochemical method was used to measure the expression of PRDX1 in tissues from 55 ovarian serous papillocarcinoma,11 borderline,21 benign ovarian tumors and 15 normal tissues.Univariate and multivariate Cox proportional hazard model were statistically assessed to investigate its association with clinicopathological factors and survival of ovarian cancer.The ovarian cancer data of 374 individuals were extracted from The Cancer Genome Atlas(TCGA ) database,we use Gene set enrichment analysis(GSEA) to further analysis.Results:PRDX1 was mainly located in the cytoplasm.The level of the expression in ovarian serous carcinoma was significantly higher in EOSC tissue than in borderline,benign ovarian tumors and normal tissues (96.36%,72.72%,57.14%,20%respectively) (P<0.05),PRDX1 showed significantly increased expression in ovarian serous carcinoma with increasing FIGO stage(P<0.05),PRDX1 is an independent factor for poor prognosis of ovarian serous carcinoma(P<0.05).Gene set enrichment analysis found that Low expression of PRDX1 was correlated with several molecular pathways of Wnt signaling pathway,ErbB signaling pathway,Notch signaling pathway,pathways in cancer,focal adhesion related to tumor.High expression of PRDX1 was enriched into oxidative phosphorylation gene set.Conclusion:Our study demonstrate that expression of PRDX1 was associated with tumorigenesis and prognosis of ovarian cancer,it might be a potential prognostic biomarker in OV patients.
