摘要
目的:观察地耳草总黄酮对肾纤维化相关因子的影响,探究地耳草总黄酮治疗肾纤维化的作用及其机制。方法:采用5/6肾切除建立大鼠慢性肾衰竭(Chronic Renal Failure,CRF)模型,ELISA检测血清促纤维化细胞因子碱性成纤维细胞生长因子(Basic Fibroblast Growth Factor,bFGF),肿瘤坏死因子-α(Tumor Necrosis Factor,TNF-α)及致炎因子白介素-2(Interleukin-2,IL-2);免疫组化检测肾脏组织胶原-Ⅰ(Collagen-Ⅰ,Col-Ⅰ)。结果:地耳草总黄酮各剂量组IL-1显著高于模型对照组(P<0.01);地耳草总黄酮高、中剂量组IL-2显著低于模型对照组(P<0.01);地耳草总黄酮各剂量组bFGF和TNF-α含量显著低于模型对照组(P<0.01);地耳草总黄酮高中剂量组Col-Ⅰ低于模型对照组(P<0.05)。结论:地耳草总黄酮通过抑制促纤维化相关因子bFGF和TNF-α和炎症细胞因子IL-2的分泌,降低Col-Ⅰ的表达,从而降低炎症反应,抑制系膜和基质的增生,改善肾小管病变,进而缓解肾纤维化。
Objective: To explore the effects and mechanism of total Herba Hyperici Japonici flavonoids in treating renal fibrosis. Methods: CRF model was induced by 5/6 nephrectomy, and IL-1, bFGF, TNF-α, IL-2 were tested was detected by ELISA (enzyme linked immunosorbent assay) , and Col-I detected by immunohistochemistry. Results: IL-1 of Total Herba Hyperici Japonici flavonoids treated groups was significantly higher while bFGF and TNF-α lower than vehicle control group (P 〈 0.01 ) . IL-2 of high and medium dose groups was significantly lower than vehicle control group (P 〈 0. 01 ), and Col-I lower than vehicle control group (P 〈 0.05) . Conclusions: Total Herba Hyperici Japonici flavonoids could relieve renal fibrosis by reducing bFGF, TNF-α, IL-2, and Col-I ; and increasing IL-1.
