摘要
目的研究金钗石斛对慢性疲劳综合征(CFS)大鼠行为学的改善作用。方法将66只SD雄性大鼠随机分成正常对照组(N组)、模型组(P1组)、造模+金钗石斛低剂量组(DF组)、造模+金钗石斛中剂量组(DM组)、造模+金钗石斛高剂量组(DH组),每组各11只。采用睡眠剥夺+力竭游泳法进行造模。造模成功后,DL组、DM组、DH组分别给予1、3及5 g/kg金钗石斛水煎液灌胃,N组与P1组灌等体积蒸馏水,1次/d,持续12 d。灌胃结束后第2天进行Morris水迷宫实验,检测相关指标。结果在Morris水迷宫实验中,P1组潜伏期明显高于N组(P<0.05),与P1组相比,DL组、DM组、DH组潜伏期明显缩短(P<0.05);在空间探索实验中,与N组比较,P1组穿越平台次数明显减少,DL组、DM组、DH组与P1组比较,穿越平台次数明显增加(P<0.05),DH组与DL组比较,穿越次数明显改善(P<0.05);有效区域运动距离实验中,P1组与N组比较,有效运动距离明显减少(P<0.05),而DL、DM、DH各组较P1组有效运动距离明显增加(P<0.05),DM、DH组与DL组比较,有效运动距离有所改善(P<0.05)。结论金钗石斛可改善CFS大鼠体能状态、学习记忆能力及行为学,缓解疲劳症状。
Objective To study the effects of Dendrobium nobile on the behavior of chronic fatigue syndrome( CFS) rats. Methods 66 SD male rats were randomly divided into five groups( each group of 11 rats),including negative control group( N group),model group( P1 group),Dendrobium nobile intervention group in dosage of low,medium and high( marked as DL,DM and DH group). Sleep deprivation and exhaustive swimming were used to establish the model. DL,DM and DH group received intragastric administration of 1,3 and 5 g/kg Dendrobium decoction respectively once a day for 12 days. N group and P1 group were administrated equal volume of distilled water. On the second day after administration,the Morris water maze test was performed,and the related indexes were detected. Results In the Morris water maze experiment,the escape latent period of P1 group was significantly longer than N group( P 〈 0. 05). Compared with P1 group,the escape latency of DL,DM and DH group was shortened( P 〈 0. 05). The results of the space exploration experiment indicated that the times of crossing the platform was obviously decreased in P1 group. Compared with P1 group,the times were increased in DL,DM and DH group( P 〈 0. 05). The effect of the intervention on DH group was better than on DL group( P〈 0. 05). For the exercise distance within range,it was the shortest in P1 group among all groups( P 〈 0. 05).And the distances were farther in DL,DM and DH group than in P1 group( P 〈 0. 05). Conclusion Dendrobium nobile could improve physical state,learning and memory ability,and behavior of CFS rats,and alleviate the symptoms of fatigue.
