摘要
检测乳及乳制品菌落总数,为达到菌落在培养基中清晰可辨,在平板计数琼脂培养基中添加2,3,5-三苯基氯化四氮唑(TTC)。以灭菌乳、发酵乳、冰淇淋为样品基质,分别加入适当菌含量的大肠埃希氏菌、金黄色葡萄球菌、粪肠球菌、枯草芽胞杆菌工作菌悬液,制备适当浓度的TTC平板计数琼脂,同时以不添加TTC的平板计数琼脂作为对照,对同一样品进行菌落计数并同国标方法结果进行对比。TTC平板计数琼脂终浓度在0.001%-0.002%时能够使菌落显色,且与不添加TTC的对照组对比结果一致。在检测成分复杂的乳及乳制品样品时添加适宜浓度的TTC,有助于菌落的辨认,能够提高检验人员菌落计数的准确性,可用于检测乳及乳制品中菌落总数。
In order to achieve the clear colony in the medium,we detect the total number of colonies of milk and dairy products by adding 2,3,5-Triphenyltetrazolium chloridechloride (TTC) into plate count agar culture medium supplemented. Compared with the national standard method, we use the same samples. This experi-ment used sterilization milk, fermented milk, ice cream as the sample matrix, and added appropriate content of Escherichia coli, Staphylococcus aureus, Enterococcus faecalis, bacillus coli bacteriasuspensionrespectively, which is aim to the preparation of appropriate concentration of TTC Plate Count Agar. At the same time, the plate count agar without TTC was used as the control groups in this test. The concentration of TTC plate count agar between 0.001%and 0.002%, can make the colony color, and the results were consistent with the control group which did not add TTC. The appropriate concentration of TTC indicator was applied to the detection of total bacterial count in milk and dairy products, which is not only helpful to identify the colony, but also can improve the accuracy of the count of the colony count.
