摘要
目的:建立高效液相色谱法检测干血滤纸片中卡马西平血药浓度的方法。方法:将30μL含卡马西平的全血滴在滤纸上,室温下晾干至少3 h,待干透后从滤纸上打孔取下6 mm直径的血斑,放入1.5 mL普通离心管中,加入含有内标的甲醇溶液200μL,超声水浴10 min,涡旋1 min,15 000 r·min^(-1)离心5 min,取上清进样。色谱柱为AgilentTC-C_(18)柱(250 mm×4.6 mm,5μm),流动相为甲醇-水(55∶45),内标为苯妥英,流速为1.0mL·min^(-1),检测波长为235 nm。结果:卡马西平血药浓度在1.0~32.0μg·mL^(-1)范围内与峰面积呈良好线性关系(r=0.999 0),方法回收率为101.3%~110.5%,提取回收率为59.8%~62.1%。结论:本方法操作简单、准确、可靠,适用于测定干血滤纸片中卡马西平的血药浓度。
OBJECTIVE To develop a method for determination of carbamazepine concentration on DBS by HPLC.METHODS A volume of 30μL blood containing carbamazepine was spotted on the filter paper and was dried at room temperature at least for 3 h,then a 6 mm diameter of disk was punched out from the center of dried blood spots into a 1.5 mL centrifuge tube,followed by adding 200μL of methanol contain internal standard.After 10 min in ultrasonic bath,the mixture was vortexmixed for 1 min and centrifuged for 5 min.A volume of 20μL supernatant was injected into the HPLC system.The Agilent TC-C18(250 mm×4.6 mm,5μm)was used as analytical columu with mobile phase consisted of methanol and water(55∶45).The flow rate was 1.0mL·min^-1 and detection wavelength was 235 nm.RESULTS The linear ranges were 1.0-32.0μg·mL^-1 for carbamazepine on DBS.The method recovery was 101.3%-110.5%,and the extraction recovery was 59.8%-62.1%.CONCLUSION The method is simple,accurate and reliable,suitable for the determination of carbamazepine on DBS.
