加味四逆散对青少期复合应激大鼠行为学及海马与皮质中5-HT、5-HIAA含量的影响 被引量:14

Effect of Modified Jiawei Sini San on Behaviors and Contents of 5-HT and 5-HIAA in Hippocampus and Prefrontal Cortex of Adolescent Rats Stimulated by Multiple Stress

摘要 【目的】观察加味四逆散对青少期复合应激大鼠行为学及海马与前额叶皮质中五羟色胺(5-HT)及其代谢产物五羟吲哚乙酸(5-HIAA)含量的影响,探讨加味四逆散抗青少期抑郁的作用机制。【方法】将8只成熟SPF级Wistar大鼠(雌雄各半)进行配种。从每只母鼠子代中随机挑选出4只雄性和4只雌性出生后24 h的子鼠,共32只,随机分为空白组、母子分离组、复合应激组(母子分离结合慢性应激)、加味四逆散组,每组8只,雌雄各半。于出生后第56天(PND 56)进行行为学观察,并采用高效液相—荧光法检测海马及前额叶皮质中5-HT及5-HIAA含量。【结果】与空白组比较,母子分离组及复合应激组PND 56大鼠体质量、糖水偏爱度、海马及前额叶皮质中5-HT和5-HIAA含量均降低(P<0.05或P<0.01),除雄性大鼠旷场实验总路程有显著性差异(P<0.05),其余大鼠活动度、中央区路程虽减少,但差异无统计学意义(P>0.05);且复合应激组大鼠体质量,糖水偏爱度,旷场实验总路程、活动度、中央区路程,海马及前额叶皮质中5-HT及5-HIAA含量均低于母子分离组(P<0.05或P<0.01)。与复合应激组大鼠比较,加味四逆散组大鼠糖水偏爱度、海马及前额叶中5-HT和5-HIAA含量明显升高(P<0.05或P<0.01),且有升高体质量和旷场实验结果的趋势。各组内不同性别比较显示,除加味四逆散组雄性大鼠体质量高于雌性(P<0.05),其余如糖水偏爱度、旷场实验、海马及前额叶皮质中5-HT及5-HIAA含量等指标均无明显性别上的差异(P>0.05)。【结论】加味四逆散可能通过提高海马及前额叶皮质中5-HT和5-HIAA含量而改善复合应激大鼠抑郁样行为。 Objective To observe the effect of modified Jiawei Sini San(MJSS)on the behaviors and the contents of 5-HT and 5-HIAA in the hippocampus and prefrontal cortex of adolescent rats stimulated by multiple stress.Methods Eight mature SPF Wistar rats,male in half,were given mating,and then 4 male and 4 female 24-hour-old offspring rats were randomly selected from each mother rat for the breeding of next generation. Eventually,a total of 32 rats were enrolled in the study and were randomly divided into blank group, materal separation(MS)group,multiple stress group(materal separation plus chronic stress),MJSS group,8 rats in each group(male in half). The behaviors of the rats on the post-natal day(PND)56 were observed,and high performance liquid chromatography(HPLC)-fluorescence method was used to analyze the contents of 5-HT and 5-HIAA in rat hippocampus and prefrontal cortex. Results Compared with the blank control group, the body weight,sucrose preference, and the contents of 5-HT and 5-HIAA in the hippocampus and prefrontal cortex of MS group and multiple stress group were decreased on PND 56(P 0.05 or P 0.01),and the parameters of field test for the male rats showed significant difference only in the total distance(P 0.05),while rat activities and the central distance just showed a declining trend(P 0.05);the above observation indexes in the multiple stress group were lower than those in the MS group(P 0.05 or P 0.01). Compared with the multiple stress group,the sucrose preference and the contents of 5-HT and 5-HIAA in the hippocampus and prefrontal cortex of MJSS group were increased(P 0.05 or P 0.01),and the body weight and field test results showed an increasing trend in MJSS group. The results of intra-group sex comparison showed that except that the body weight of male rats was higher than that of the female rats in MJSS group(P 0.05),but no significant differences of the sucrose preference, field test results,or the contents of 5-HT and 5-HIAA in the hippocampus and prefrontal cortex were shown between genders of other groups(P 0.05). Conclusion MJSS can improve the depression behaviors of adolescent rats stimulated by multiple stress through increasing the contents of 5-HT and 5-HIAA in the hippocampus and prefrontal cortex.
