正交试验法优选莲子心总生物碱栓剂制备工艺 被引量:1

Optimization on preparation procedure of total alkaloids extracted from lotus plumule suppository by orthogonal experiment

摘要 目的优选莲子心总生物碱栓剂的制剂处方。方法以硬脂酸聚烃氧(40)酯和聚乙二醇2000为主要基质,甘油为保湿剂,吐温-80为表面活性剂,采用正交试验法筛选莲子心总生物碱栓剂的制剂处方,以栓剂外观及30 min体外溶出度的综合评分为指标评价栓剂质量。同时考察脱模时间及成型温度对栓剂质量的影响。结果优选出的最佳处方组成为硬脂酸聚烃氧(40)酯∶聚乙二醇2000∶甘油(7.5∶2∶1.5),吐温-80最适宜的含量为1.5%,最佳脱模时间为30 min,成型温度为70℃。结论优选出的制剂工艺设计合理可行,外观完整光滑,体外释药迅速。 Objective To optimize the formulation of total alkaloids extracted from lotus plumule (TLAP) suppository. Methods The TLAP suppository was prepared by using S-40 and PEG-2000 as the main base, glycerine as humectant and tween-80 as a suffactant. Suppository's appearance and 30 min's dissolution rate in vitro were used as the indexes to evaluate the quality of the suppository, orthogonal design test was employed to select the optimum prescription. Meanwhile, the impact of stripping time and molding temperature on the quality of TLAP suppository were also investi- gated. Results The best prescription of TLAP suppository was S-40: PEG-2000:glyeerol at 7.5:2:1.5, and the most suit- able content of tween-80 was 1.5%. The optimal stripping time was 30 minutes and molding temperature was 70℃. Conclusion The optimized prescription is rational and practicable. The prepared suppository appearance is smooth and complete, rapid in vitro release.
