温针灸足三里、肾俞对类风湿性关节炎模型RhoA蛋白表达的影响 被引量:24

Effects of Warm Acupuncture on the Expression of Protein RhoA in Rheumatoid Arthritis Rats

摘要 目的探讨温针灸对类风湿性关节炎模型大鼠RhoA(Ras homolog gene family,member A)蛋白表达影响的实验研究。方法清洁级健康3月龄SD大鼠60只(雌雄各半),随机分为正常组、模型组和温针灸组三组,每组20只,采用皮内注射牛II型胶原接种诱发制备大鼠类风湿性关节炎模型。温针灸组于造模第1天取足三里和肾俞穴温针灸治疗,采用手针,刺激量以大鼠耐受为度,1次/d,共21d。所有大鼠于相应处理结束后,采用排水法测大鼠膝关节部体积,放免法检测血清白介素含量,免疫印迹法检测膝关节滑膜组织RhoA蛋白的表达。结果与正常组比较,模型组大鼠膝关节肿胀明显,血清中白介素含量较高,膝关节滑膜组织RhoA蛋白表达增加,差异具有统计学意义(P<0.01);与模型组比较,温针灸组大鼠膝关节肿胀改善明显,白介素含量减少,RhoA蛋白表达均降低,差异具有统计学意义(P<0.05)。结论温针灸能有效调节RA大鼠关节炎性损伤状况,降低RA大鼠血清白介素和骨关节滑膜组织RhoA蛋白含量,这可能是温针灸通过调节RhoA蛋白改善RA的调节机制之一。 Objective To determine the effect of warm acupuncture (WA) on the expression of protein RhoA in rheumatoid arthritis (RA) model rats. Methods Up to 60 3 - month old Sprague - Dawley (SD) rats were randomly divided into normal, model and WA groups, 20 rats in each group. Rats were prepared as RA models by administering intradermal injection of bovine type II collagen emulsified in incomplete Freund' s adjuvant for 21d. Zusanli ( ST 36) and Shensbu ( BL 23 ) were applied for treatment of the WA group starting on the first day, with manual acupuncture and moxibustion therapy, and evaluation in rats pain tolerance, once a day for totally 21d. Rat knee joint volume was measured by drainage method, contents of serum interleukins were detected by radioimmunoassay while expression levels of RhoA in synovial tissue of knee joint were determined by western blotting. Results Swelling in the knee joint of rats in model group was signifi- cantly increased, serum interleukins content was higher, and the knee joint synovial tissue RhoA protein expression increased significantly while compared with those in the normal group ( P 〈 0. 01 ). These parameters were lower in WA group than those in model group ( P 〈 0. 05 ). Conclusion WA can effectively improve the articular inflammatory damage situation of RA rats, reduce the content of serum interleukins and protein RhoA in synovial tissue, this may be a useful mechnism of WA to reduce disease severity in collagen - induced arthritis by regulating RhoA.
