摘要
目的探究宁心合剂壳聚糖絮凝澄清最佳工艺。方法采用壳聚糖絮凝法对于宁心合剂进行除杂处理,以黄芪甲苷、(R,S)-告依春、丹酚酸B、总多糖保留率为评价指标,采用单因素考察提取液浓度、壳聚糖加入量、药液pH值、保温时间、搅拌时间对壳聚糖絮凝澄清工艺的影响。在其基础上利用正交试验考察提取液浓度、壳聚糖加入量、药液pH值对絮凝效果的影响,并将絮凝法与醇沉法、滤过法进行比较。结果宁心合剂的最佳壳聚糖絮凝工艺为以提取液的生药浓度为0.75g·ml^(-1),药液pH=6,絮凝温度为40℃,壳聚糖加入量为1g·L^(-1),快速搅拌15min。结论可以将壳聚糖絮凝作为宁心合剂的澄清工艺。
Objective To optimize flocculation clarification technology conditions of Ningxin oral liquid by chitosan. Methods Using chitosan clarify Ningxin oral liquid, taking absorbance and retention rate of Astragaloside IV, Epigoitrin, Salviandic acid B and total polysaccharide, using single factor investigation to check the potency of oral liquid, the dosage of chitosan, pH, soaking time and mixing time. On its foundation, using orthogonal test to check the potency of oral liquid, the dosage of chitosan, pH, and compare the effect of clarification of alcohol - precipitation and filtering technology. Results The best chitosan clarification is that the potency of oral liquid is 0.75 g · mL^-1, pH = 6, the soaking time is 40℃, the dosage of chitosan is 1 g ·L^-1, the mixing time is 15min. Conclusion Chitosan flocculation method could be used as clarification technology of Ningxin oral liquid.
