摘要
在体外研究结果表明,乳源性小肽DELQ具有良好的生物活性基础上,采用秀丽线虫为实验模型,研究DELQ的抗衰老功效。首先以生殖能力为指标,在线虫培养基中添加不同浓度的DELQ,选出最佳作用浓度;同时设立对照组和实验组,评价DELQ对线虫寿命以及生殖能力的影响。然后采用热应激和氧化应激实验,探讨DELQ的抗衰老机理。结果表明:400 mg/L的DELQ为最佳作用质量浓度;DELQ在一定质量浓度下能延缓线虫衰老,显著增加SOD质量浓度;提升线虫生殖能力;耐热力增加不明显;但能显著提高线虫的抗氧化力,MDA及ROS值均下降,SOD明显增高。因此,乳源性小肽DELQ能通过清除自由基,提升机体抗氧化力来延缓线虫衰老,故推测DELQ具有一定的抗衰老功效。
This research was based on the vitro study that milk-derived peptide DELQ showed good biological activities, c. elegans were used as experimental model to study the anti-aging efficacy of DELQ, which were treated with different concentrations of DELQ, tested by reproductive capacity assay to determine the effective dosage. The synchronized L4s were treated as control group and experimental group, in which the indexes of lifespan, fecundity, acute heat and oxidative stress were included, to explore the initial mechanism of anti-aging func- tion. As results showed, 400 mg/L of DELQ was the best dosage for c. elegans to extend lifespan and enforce fecundity; DELQ had a signifi- cant effect on c. elegans' bodies in the level of SOD, MDA and ROS to protect against oxidant stress, but not did so in heat resistance. In conclusion, DELQ performed well in scavenging free radicals and enhancing oxidation resistance to help extend lifespan in some extent.
