摘要
目的探讨塞来昔布(Celecoxib)对压力超负荷性心肌肥厚大鼠模型心室结构功能及前列环素(prostaglandin I2,PGI2)、血栓烷A2(thromboxane 2,TXA2)含量的影响。方法将雄性Wister大鼠随机分为假手术组、模型组、Celecoxib 2周组及Celecoxib 4周组。超声心动图测量心功能各项指标。光镜下左心室组织切片,测量左心室质量指数(left ventricular mass index,LVMI),ELISA法检测血浆及心肌组织中PGI2、TXA2的含量。结果模型组、Celecoxib 2周组、Celecoxib 4周大鼠Tie指数高于假手术组,Celecoxib 2周组和Celecoxib 4周组Tie指数低于模型组(P<0.05)。模型组和Celecoxib 2周组IVRT高于假手术组,Celecoxib 4周组IVRT低于模型组和Celecoxib 2周组(P<0.05)。模型组、Celecoxib 2周组、Celecoxib 4周组射血分数(ejection fraction,EF)、左心室短轴缩短率(fraction shortening,FS)低于假手术组,CO高于假手术组(P<0.05)。Celecoxib 4周组血浆中PGI2含量高于假手术组,模型组、Celecoxib 4周组心肌组织中PGI2含量高于假手术组和Celecoxib 2周组(P<0.05)。2组血浆和心肌组织中TXA2含量差异无统计学意义(P<0.05)。结论心肌肥厚早期心脏整体功能及心室结构出现明显变化,随时间的延长,心室收缩功能也出现障碍;心肌肥厚大鼠血浆和心肌组织中PGI2发生变化;Celecoxib可以减轻心肌肥厚程度,但长期使用可能产生不良效果。
Objective To investigate the effect of Celecoxib on the function and structure of the ventricular,and on the content of prostaglandinI2(PGI2)and thromboxane2(TXA2)in models of rats with pressure-overload myocardial hypertrophy.Methods Female Wister rats were randomly divided into sham operation group,model group,Celecoxib 2 weeks group and Celecoxib 4weeks group.Echocardiographic measurements were used to measure cardiac function indexes.Left ventricle tissue sections were viewed by light microscope.LVMI was measured and content of PGI2,TXA2 were measured by ELISA assay in plasma and myocardial tissue.Results In model group,Celecoxib 2 weeks group and Celecoxib 4 weeks group,Tie indexes were higher than that in the the sham group.In Celecoxib 4 weeks group and 2 weeks group,Tie indexes was lower than that in the model group(P〈0.05).In model group and Celecoxib 2weeks group,IVRT was higher compared with the sham group.IVRT of Celecoxib 4 weeksgroup was lower than that in the model group and Celecoxib 2weeks group(P〈0.05).In model group,Celecoxib 2weeks group and Celecoxib 4weeks group,EF,FS were lower than that in sham group,CO was higher than that in sham group(P〈0.05).The content of plasma PGI in Celecoxib 4 weeks group was higher than that in the sham group and model group.PGI2 in myocardial tissue of Celecoxib 4weeks group was higher than that in sham group and Celecoxib 2weeks group(P〈0.05).There was no significant difference in the content of TXA2 in plasma and cardiac tissue between the 2 groups(P〈0.05).Conclusion At the early stage of cardiac hypertrophy,ventricular function and structure were significantly changed.With the extension of time,ventricular systolic function also appeared obstacles.PGI2 in plasma and myocardial tissue of cardiac hypertrophy rat was changed.Celecoxib can alleviate cardiac hypertrophy.However,long-term use may have a side effect.
