摘要
目的:建立高效液相色谱法(HPLC)法测定银黄降脂袋泡茶中绿原酸和大黄素含量的方法。方法:采用DiamonsilC18柱,绿原酸和大黄素分别以乙腈-0.4%磷酸水(13∶87)和甲醇-0.1%磷酸水(80∶20)为流动相,以327nm和254nm为检测波长,流动相流速为1.0mL/min;柱温为30℃。结果:绿原酸和大黄素的线性回归方程分别为ˇy=17.955x-0.2848,相关系数r=0.9999,线性范围为0.041~0.41μg;ˇy=33.975x+186.36,相关系数r=0.9995,线性范围0.0395~0.395μg。平均加样回收率分别100.38%(RSD为2.34%,n=9)和101.21%(RSD为2.59%,n=9)。结论:该方法简便、快速、重现性好、结果准确,可用于银黄降脂袋泡茶的质量控制。
Objective:To establish the high performance liquid chromatography(HPLC)method for the determination of chlorogenicacid and emodin in Yinhuang Jiangzhi Tea.Methods:The contents of chlorogenicacid and emodin in Yinhuang Jiangzhi Tea were determined with Diamonsil C18 column by elution of acetonitrile-0.4%phosphoric acid in water(13:87)and methanol-0.1%phosphoricacidin water(80:20),at 327 nm and 254 nm respectively.The flow rates were 1.0mL/min,and the column temperatures were 30℃.Results:The correlation between Injection Volume and peak area of chlorogenic acid and emodin were Y=17.955X-0.2848(R=0.999 9,0.041-0.410μg)and Y=33.975X+186.36(R=0.999 5,0.0395-0.3950μg),respectively.The mean recovery of chlorogenic acid and emodin were 100.38%(RSD=2.34%,n=9)and 101.21%(RSD=2.59%,n=9),respectively.Conclusions:The HPLC methods are relatively simple,accurate,and specific,and can be used for the quality control of Yinhuang Jiangzhi Tea.
