半滑舌鳎鳃黏液免疫相关酶活性对黄芪多糖的免疫应答 被引量:15

Immune Response of Activity of Enzymes Related to Immunity to Astragalus Polysaccharides in Gill Mucus of Semi-smooth Sole Cynoglossus semilaevis

摘要 在水温25℃下,给体质量(5.8±1.5)g的半滑舌鳎投喂在基础饲料中添加0、300、600、900、1200、1500mg/kg和1800mg/kg黄芪多糖的饲料后,每隔18d检测半滑舌鳎鳃黏液溶菌酶、过氧化物酶、碱性磷酸酶和酸性磷酸酶的活性,以检测半滑舌鳎鳃黏液对黄芪多糖的免疫应答及较佳剂量。90d的免疫结果显示,半滑舌鳎鳃黏液溶菌酶、过氧化物酶和碱性磷酸酶活性先增强后降低;半滑舌鳎鳃黏液酸性磷酸酶活性对低剂量(300~600mg/kg)和中高剂量(1200~1800mg/kg)黄芪多糖的免疫应答,随试验时间延长,分别呈增强及先增强后减弱的变化趋势。试验结束时,中低剂量(600~900mg/kg)黄芪多糖组半滑舌鳎鳃黏液溶菌酶、过氧化物酶、碱性磷酸酶和酸性磷酸酶活性均极显著高于对照组(P〈0.01)。从成本、高非特异免疫活性的角度看,半滑舌鳎配合饵料中黄芪多糖较佳添加剂量为600mg/kg。 Juvenile semi-smooth sole(Cynoglos sussemilaevis)with body weight of(5.8±1.5)g was fed basic diet supplemented with astragalus polysaccharides(APS)at a dose of 0,300,600,900,1200,1500 and 1800mg/kg diet with triplication at water temperature of 25℃for 90 d,and the activities of lysozyme(LSZ),peroxidase(POD),alkaline phosphatase(AKP)and acid phosphatase(ACP)in the gill mucus were detected in the sole fed the diets containing various doses of APS in a 18 day interval to investigate the immune response of activities of LSZ,POD,AKP and ACP in the gill mucus to APS and further explore the optimal dietary dose of APS.The activities of LSZ,POD and AKP were found to be first increased and then decreased in the gill mucus.The activity of ACP was shown to be elevated with elapse of APS immunity in the low dose of APS groups(300—600mg/kg),and to be increased first and then decreased in the middle and high levels of APS groups(1200—1800mg/kg).At the end of the experiment,there were very significantly higher activities of LSZ,POD,AKP and ACP in the gill mucus in the low and middle dose APS groups(600—900mg/kg)than those in the control group(P〈0.01).It is recommended that the optimal dietary dose of APS be 600mg/kg diet for semi-smooth sole juveniles based on feed cost and immune efficiency.
