摘要
目的探讨健脾养血祛风方对脾虚证慢性湿疹小鼠模型的作用机制。方法将50只健康雄性小鼠随机分为空白组、模型组、西药组、中药高剂量组及中药低剂量组。除空白组外,其余小鼠均制备脾虚证慢性湿疹模型。中药高、低剂量组给予相应剂量健脾养血祛风方干预,西药组给予盐酸左西替利嗪干预,空白组和模型组均给予等量生理盐水。灌胃14d后观察各组小鼠皮肤组织病理变化情况、耳肿胀度(耳质量差)、耳厚度差、脾脏指数及胸腺指数。结果中药高、低剂量组以及西药组小鼠病变皮肤组织病理示表皮角化过度不明显,棘层肥厚明显减轻,真皮内炎性浸润显著减少,内皮细胞肿胀减轻。与空白组比较,其他各组小鼠耳厚度差及耳肿胀度均显著增加(P均<0.05);各给药组小鼠耳厚度差及耳肿胀度均明显低于模型组(P均<0.05),各给药组间比较差异无统计学意义(P均>0.05)。与空白组比较,其他各组小鼠脾脏指数及胸腺指数均显著升高(P均<0.05);各给药组小鼠脾脏指数及胸腺指数均明显低于模型组(P均<0.05),各给药组间比较差异无统计学意义(P均>0.05)。结论健脾养血祛风方能够显著减轻脾虚证慢性湿疹小鼠模型耳肿胀度,减少耳厚度差;下调脾虚证慢性湿疹小鼠模型增高的脾脏指数及胸腺指数;抑制病变组织炎性反应,改善慢性湿疹皮损的病理结构。该方对脾虚证慢性湿疹小鼠模型具有较好的抗炎、抗过敏作用。
Objective It is to investigate the possible therapeutic mechanism of Jianpi Yangxue Qufengfang (JPYXQFF) on mice model of syndrome of spleen deficiency with chronic eczema. Methods Fifty healthy male mouse were randomly divided into blank group, model group, western medicine group, TCM high dose group and TCM low dose group. Except blank group, all groups were induced to models of syndrome of spleen deficiency with chronic eczema. The animals were intervened with corresponding dose of JPYXQFF in TCM high and low :group, and with levocetirizine hydruchloride in western medicine group, with equal dose of physiological saline in blank and model group. The pathological changes of skin, ear swelling degree, ear thickness difference, spleen index, thymus index were observed after 14 days of intervention. Results TCM high and low dose group and western medicine group had obvious inhibitory effect on the local inflammation, and can improve the pathological of skin lesions. There was significant difference between the blank group and the others in ear swelling degree and ear thickness. ( all P 〈 0.05) ; the ear swelling degree and ear thickness difference in all treatment group were significantly thinner than that in the model group ( all P 〈 0.05 ) , but there was no significant difference among the treatment groups ( all P 〉 0.05 ). The spleen index and thymus index were significantly increased in the other groups compared with the blank group ( all P 〈 0.05) , and those indexes were decreased in the treatment groups compared with the model group ( all P 〈 0. 05 ) , but there was no significant difference among the treatment groups ( all P 〉 0. 05 ). Conclusion JPYXQF can significantly reduce the ear swelling degree and ear thickness difference of mouse model of spleen deficiency syndrome with chronic eczema, and decrease the spleen index and thymus index, inhibit the inflammatory reaction of the tissue lesions, improve pathological structure of chronic eczema lesions. It has better anti - inflammatory and anti allergic effects on the mouse model of spleen deficiency syndrome of chronic eczema.
