复方酮康唑喷膜对豚鼠湿疹模型的实验研究 被引量:13

摘要 目的 观察复方酮康唑喷膜对豚鼠湿疹模型的疗效。方法 选择健康雌性豚鼠25只,每只背部剃毛4处,应用卵清蛋白(OVA)为抗原,激发豚鼠皮肤变态反应制备湿疹模型。将制备的模型随机分为A,B,C,D和E五组(N=100,n=20)。激发后给药,其中A,B,C组给予含高、中、低浓度地塞米松的复方酮康唑喷膜,D组给予地塞米松止痒霜,E组空白对照。根据形态学和组织病理学观察进行疗效评估。结果 豚鼠皮肤显示湿疹样皮损特点,组织病理呈湿疹样改变。药效学结果显示治疗1周时,A,B,C,D四组红斑、水肿评分和总分均明显降低(P<0. 05),而E组无明显变化。复方酮康唑喷膜与地塞米松止痒霜疗效一致,但给药次数明显减少。四组抑制率均在50%左右,B组抑制率最高,表明该组处方比较优化。结论 复方酮康唑喷膜能快速地抑制真皮炎症反应和变态反应性湿疹形成。 Objective To observe the effect of compound ketoconazole spraying film on guinea pig eczema model.Methods 25 female guinea pigs were shaved at four sites on the back each one. The eczema models were prepared using ovalbumin as an antigen to stimulate their shaving sites,and divided into A,B,C,D and E groups at random(N=100,n=20).After the eczema models being challenged, the compound ketoconazole spraying film was began to administrate L.A,B and C groups were administrated compound ketoconazole spraying films containing different concentration dexamethasone respectively,D group was administrated dexamethasone antipruritic cream and E group was considered as blank group.The scores criteria of morphology and histopathology were defined according to related literatures.The effect of compound ketoconazole spraying film on guinea pig eczema models was evaluated based on the scores.Results Morphology and histopathology of guinea pigs skin showed the eczema like features. While guinea pig eczema models were administrated for one week, there were significant differences for erythema,edema and total scores for A,B,C and D groups between pretreatment and post treatment ( P <0.05).Compared with the control,a significant difference could be seen for different treatment groups ( P <0.05).The outcome of pharmacodynamics showed that the compound ketoconazole spraying film had identical effect on guinea pig eczema models with dexamethasone antipruritic cream.However,compared with dexamethasone antipruritic cream, the frequencies of administration of ketoconazole spraying film decreased significantly.The inhibition rate of four treatment groups were about to 50 percent,but B group among three compound ketoconazole spraying film treatment groups was highest,which showed that the prescription of this group was better than the others.Conclusion The compound ketocozanole spraying film can quickly inhibit dermal inflammatory reaction and allergic eczema formation.
