Preparation of human hair protein solution by alkali-reduction method

摘要 采用碱-还原法溶解废弃人发制备人发蛋白液,通过氢氧化钠用量、亚硫酸钠用量、反应温度和反应时间的单因素对比试验,以人发溶解率和人发蛋白液质量分数为测试指标,确定最佳反应条件.结果表明:Na2SO325 g/L、Na OH 6 g/L、SDS 10 g/L、反应温度80℃、反应时间3 h时,人发溶解率达到86.73%,人发蛋白液质量分数为5.05%,且稳定性好.SDS-PAGE显示人发蛋白分子质量主体分布在37 kDa以下.FTIR显示,人发蛋白的分子结构从α-螺旋结构转变为β-折叠结构. The alkali-reduction method was used to dissolve waste human hair for preparing protein solution. Taking the dissolution rate of human hair and the mass fraction of protein solution as test indexes, the op- timum process conditions were determined by varying the dosage of sodium hydroxide and sodium sulfite, re- action temperature and reaction time. The results showed that the dissolution rate was up to 86.73% and the mass fraction was 5.05% when sodium sulfite was 25 g/L, sodium hydroxide was 6 g/L, SDS was 10 g/L and reacted at 80 ℃ for 3 h. SDS-PAGE analysis showed that the molecular weight distribution of protein was below 37 kDa. FITR analysis indicated that the molecular structure of hair protein was transformed from α-helix to β-sheet.
