电针对CFA慢性炎性痛大鼠患侧背根神经节NR1及其丝氨酸890位点磷酸化的影响 被引量:2

摘要 [目的]观察电针(EA)对完全弗氏佐剂(CFA)诱导的慢性炎性痛大鼠患侧背根神经节(DRG)N-甲基-D-天门冬氨酸受体亚基1(NR1)受体及NR1丝氨酸890位点磷酸化(p NR1 serine-890)的影响。[方法]健康雄性SD大鼠30只,随机分为正常对照组(N组),模型对照组(CFA组),电针组(EA组),每组10只。右后足底注射CFA 0.1ml以建立慢性炎性痛模型,分别测量大鼠造模前,造模后第1d、3d、5d、7d、9d患侧痛阈变化。免疫荧光法检测大鼠患侧DRG中NR1阳性细胞表达率,免疫印迹法检测大鼠患侧DRG中p NR1 serine-890的表达量。[结果]CFA造模后1d,CFA组与EA组足缩腿阈(PWTs)均低于N组(P<0.01),CFA组与EA组差异无统计学意义;治疗3d起,EA组PWTs呈现升高趋势,于第9d明显高于同时点CFA组(P<0.01)。CFA造模后第9d,与N组比较,CFA组大鼠患侧NR1和p NR1表达均有显著上升(P<0.01,P<0.01);与CFA组比较,EA组大鼠患侧NR1和p NR1表达均下降(P<0.01,P<0.01)。[结论]电针能有效拮抗CFA诱导的大鼠慢性炎性痛,此作用可能与电针下调大鼠患测DRG中的NR1表达量以及减少NR1丝氨酸890位点磷酸化相关。 [Objective]To observe the effect of electroacupuncture(EA) on N-methyl-d-aspartate receptors I(NR1) and phosphorylationin of NR1 at serine- 890 site in ispilateral dorsal root ganglion(DRG) of rats with chronic inflammatory pain induced by CFA. [Methods]30 adult male Spragne Dawley(SD) rats were randomly divided into 3 groups:normal(N) group, CFA group, EA group, each group 10 rats. The model of chronic inflammatory pain was established by subcutaneously injecting CFA(O.lml per rat) into the fight hind paw. The paw withdrawal thresholds(PWTs) were measured before CFA injection, as well as at ld, 3rd, 5th, 7th,9th day after CFA injection. The expression of NR1 in ispilateral DRG was detected by immunofluorescence assay and the expression of pNR1 serine-890 in ispilateral DRG was detected by western blot. [Result]At ld post-injection, the PWTs of CFA group and EA group were significantly lower than N group(P〈0.01), the differences between CFA group and EA group didn't have statistical significance. Compared with CFA group, PWTs of EA group showed increasing tendency from 3d post-injection and were obviously higher than CFA group at same time point(P〈O.01). Compared with N group, the expression of NR1 and pNR1 serine-890 in ispilateral DRG of CFA rats both increased with obvious significance(P〈0.01, P〈 0.01).Compared with CFA group, expression of NR1 and pNR1 serine-890 in ispilateral DRG of EA rats both reduced with obvious significance(P〈0.01, P〈0.01). [Conclusion]EA has an obvious analgesic effect on chronic inflammatory pain induced by CFA, the effect is possibly associated with downregulation of NR1 and phosphorylation of NR1 at serine-890 site in ispilateral DRG.
