红阳猕猴桃叶片再生体系的建立 被引量:6

摘要 文章以红阳猕猴桃组培苗的叶片为外植体,研究了不同植物生长调节剂对愈伤组织的诱导、不定芽的分化及生根的影响。结果表明,MS+0.5mg/L ZT(玉米素)+0.5mg/L 2,4-D(2,4-二氯苯氧乙酸)诱导产生淡黄色疏松的愈伤组织,MS+3.0mg/L 6-BA(6-苄基腺嘌呤)+1.0mg/L NAA(萘乙酸)诱导产生绿色致密的愈伤组织,愈伤组织诱导率均为100%。在MS+5.0mg/L 6-BA+0.5mg/L NAA培养基上愈伤组织的不定芽分化率达到100%。在1/2 MS+1.2mg/L IBA(吲哚丁酸)培养基上不定芽生根率为83.3%。 Using the leaves of test-tube seedlings of Actinidia chinensis cv.Hongyang as explants,the effect of different growth regulating substances on the initiating of callus,the regeneration and rooting of adventitious buds was researched.The results indicated that the medium of MS supplemented with0.5mg/L zeatin and 0.5mg/L 2,4-dichlorophenoxyacetic acid(2,4-D)induced the faint yellow and loose callus.The medium of MS supplemented with 3.0 mg/L 6-benzylaminopurine(6-BA)and1.0mg/L naphthaleneacetic acid(NAA)induced the green and compact callus.Both of the induction efficiency achieved 100%.The maximum regeneration frequency of adventitious buds was 100% when MS was supplemented with 5.0mg/L 6-BA and 0.5mg/L NAA.The best medium for rooting of the adventitious buds was 1/2 MS medium with 1.2mg/L IBA,and the rooting rate was 83.3%.
