摘要 提出了一种新的肝素钠酶解工艺,利用2709碱性蛋白酶和胰蛋白酶组合而成的复合酶催化酶解肝素钠。以肝素总单位数为评价指标,通过单因素实验和正交设计实验得到了复合酶酶解工艺的最佳工艺参数:2709碱性蛋白酶和胰蛋白酶的酶量比以2∶1的比例组合,温度为58℃,盐浓度为0.78 mol/L,p H为9,总酶量为0.3 g/L,反应时间为2 h。与现有单一蛋白酶酶解工艺相比,肝素得率提高了6%,肝素粗品吸光度明显降低,肝素质量得到提高。复合酶酶解肝素钠的新工艺为实际车间生产提供了重要的参考。 To improve the enzymatic process of heparin sodium, a kind of innovative enzymatic technology of heparin sodium is presented. The combinative enzyme which combines 2709 alkaline protease with trypsin is adopted in enzymatic process. The optimum conditions in the enzymatic process are obtained through the single factor experiments and orthogonal experiments:2:1 of 2 709 alkaline protease and trypsin,58℃ of reaction temperature,0. 79 moL/L of salinity ,9 of pH ,0. 3 g/L of the amount of eombinative enzyme and 2 hours of the reaction time. Compared to the original single enzymatic technology, the yield of crude heparin is improved by 6% and the quality of crude heparin is also improved. The new enzymatic technology of heparin sodium by combinative enzyme provides an important reference for practical workshop production.
