摘要
目的检测鲍曼不动杆菌(Acinetobacter baumannii)质粒上Ι类整合子基因情况,以探讨质粒上Ι类整合子与鲍曼不动杆菌耐药性的关系。方法对71株MDR鲍曼不动杆菌用VITEK分析仪进行药敏分析,通过PCR检测质粒上Ι类整合酶基因及Ι类整合子的可变区并进行测序。同时我们通过质粒的接合试验探索整合子基因是否会通过质粒进行传递。结果 71株鲍曼不动杆菌均表现为多重耐药,其中有67株(94.4%)检测出I类整合子系统,I类整合子阳性菌株中,44(65.7%)株细菌携带基因盒,且主要呈现3种大小,分别为3074、2600和1700bp。整合子可通过质粒的接合试验传递给受体菌并稳定传代。结论Ι类整合子在多重耐药鲍曼不动杆菌质粒上广泛存在,可变区可捕获不同基因呈多种类型,且可通过质粒水平传播,与鲍曼不动杆菌多重耐药性之间有密切的关系。
Objective To detect the class I integron gene in A. baumannii plasmids so as to explore the correlation between class I integron in plasmids and drug-resistance of A. baumannii. Methods Drug sensitivity of the 71 Multi-drug resistant (MDR) A. baumannii were analyzed by VITEK, and class I integrase and variable regions in plasmids were explored by polymerase chain reaction (PCR) and sequencing. Simultaneously we explored if class I integron gene can transmited by conjugation. Results Totally 71 strains of A. baumannii were multi-drug resistant and class I integrase genes were observed in 67 of 71 strains of A. baumannii. In class I integron positive strains variable regions were amplified in 44 of 67 and were of 3 size mainly including approximately 3074bp, 2600bp and 1700bp respectively. The class I integron could be stably transferred to the recipient by the way of conjugation. Conclusion The class I integron were widespread in A. baumannii plasmids. Variable regions can capture different gene cassettes. We found class I integron played an important role in multi-drug resistance ofA. baumannii.
