摘要
目的 建立一种新的简便易行的AmpC酶的表型筛选方法并对其进行评价。方法 采用表型筛选和三维试验 ,对 1 4 4株阴沟肠杆菌进行AmpC酶检测 ,对检测结果进行比较 ,并检测部分菌株的等电点和ampD基因。结果 1 4 4株阴沟肠杆菌表型筛选结果显示高产AmpC酶的菌株共有35株 ,占 2 4 .31 % (35/1 4 4 )。表型筛选与三维试验相比总符合率为 89.58% (1 2 9/1 4 4 )。 2种方法检测结果相反的有 5株菌 ,其中 4株菌的等电点条带均不能被氯唑西林抑制 ,另 1株菌株等电点检测有 5条带 (其中 2条被氯唑西林抑制 )。 3株ampD基因阳性菌株测序结果显示均存在 71 ,72 ,75位氨基酸发生改变 ,其中有 2株还存在 1 0 1位氨基酸改变。结论 表型筛选法检测AmpC酶结果可靠、方法简便 。
Objective To set up and evaluate a new convenient phenotype screening method to detect AmpC enzyme. Methods Phenotype screening and three dimensional tests were performed to detect the 144 strains of Enterobacter cloacae. AmpD gene and isoelectric points were detected in part of strains. Results The results of phenotype screening showed 35 strains (24.31%) highly produced AmpC enzymes. Phenotype screening compared with three dimensional test, the coincidence rate was 89.58% (129/144) and 5 strains had opposite results. Isoelectric point straps of 4 strains were not inhibited by cloxacillin. The other strain had 5 isoelectric point straps which 3 straps were inhibited by cloxacillin. The ampD gene sequencing results of 3 strains showed there were amino acid alterations in position 71, 72, 75 and 101. Conclusions Phenotype screening is reliable to detect AmpC enzyme and easy to use in clinical laboratories.
