纯镁超声微弧氧化载牛乳铁蛋白复合膜层植入体的生物相容性 被引量:1

Biocompatibility of pure magnesium ultrasonic micro-arc oxidized bovine lactoferrin composite membrane implant

摘要 目的探讨纯镁超声微弧氧化膜层载入牛乳铁蛋白,对其骨结合能力的影响。方法以纯镁微弧氧化(Microarc oxidation,MAO)(a)组为对照组,实验组分别是纯镁微弧氧化-硅烷(b)组,纯镁微弧氧化-硅烷载牛乳铁蛋白(c)组,分别分组植入大白兔下颌骨内。在不脱钙的情况下,通过扫面电镜观察植入后膜层的形貌,锥形束投照计算机重组断层影像设备(Cone beam CT,CBCT)观察镁基体与骨组织结合及其降解情况,甲苯胺蓝染色观察骨组织形成形态,采用能谱分析仪测定不同涂层的表面元素的情况。结果通过CBCT的观察其骨组织愈合较好的是8周的(c)组,(a)组较(b)组差,甲苯胺蓝染色骨组织生长的统计学分析,均表现出(c)组优于(b)组,(b)组优于(a)组。结论纯镁超声微弧氧化以硅烷为偶联载牛乳铁蛋白复合膜层,使镁基体具有更强促进骨结合的能力,增强生物相容性。 Objective To investigate the effect of pure magnesium ultrasonic micro-arc oxidized filmcontaining bovine lactoferrin on the binding ability. Methods Microarc oxidation(MAO) processed group was used as the control group(Group(a)). One experimental group was processed by pure magnesium-micro-arc oxidation with additional silane solution soaking(group(b)) and another experimental group was processed by pure magnesium-micro-arc oxidation with additional silane solution and lactoferrin soaking(group(c)). They were implanted into the mandible of white rabbits respectively. In the absence of decalcification,the morphology of the membrane after implantation was observed by scanning electron microscopy. Cone beam computed tomography(CBCT) was used to observe the combination of magnesium matrix and bone tissue. Degradation,toluidine blue staining was used to observe the morphology of bone formation,and the surface elements of magnesium matrix of different coatings were determined by energy spectrum analyzer. Results According to CBCT results,the bone tissue healing was best for group(c) in 8 weeks and group(b) was better than group(a). statistical analysis of toluidine blue staining showed that group(c) was better than group(b),and group(b) was better than group(a). Conclusions Pure magnesium ultrasonic micro-arc oxidation with silane as the coupling of bovine lactoferrin composite film layer improves bone bonding and enhances biocompatibility.
