摘要
目的:观察蓝布正提取物对阿尔茨海默症(AD)模型小鼠学习记忆能力及相关蛋白表达的影响。方法:将54只KM小鼠随机分为6组:正常对照组、模型对照组、阳性对照组、蓝布正高、中、低剂量组。除正常对照组注射等体积生理盐水外,其余各组小鼠每天皮下注射D-半乳糖联合腹腔注射亚硝酸钠复制AD模型小鼠。造模的同时灌胃相应溶媒,除正常对照组和模型对照组灌胃等体积蒸馏水外,其余各组灌胃相应药物,连续60 d。后行水迷宫检测小鼠的逃避潜伏期和穿越平台次数。水迷宫结束后,麻醉小鼠取脑组织,免疫组织化学法检测脑组织TNF-α、AKT的表达。结果:与正常对照组比较,模型对照组逃避潜伏期明显延长,穿越平台的次数显著减少,TNF-α、AKT的表达显著增高;蓝布正各剂量组较模型组上述指标明显改善。结论:蓝布正对D-半乳糖联合亚硝酸钠复制的AD小鼠有改善作用,能下调炎症因子的表达,增强老年痴呆小鼠的学习记忆能力。
Objective To observe the effect of Gei Herba extract on learning and memory ability and related protein expression in alzheimer's disease (AD)model mice.Methods 54 KM mice were randomly divided into 6 groups:Normal control group,model control group,positive control group,blue cloth high,medium and low dose group.In addition to normal control group,other mice in each group were subcutaneously injected with d -galactose and intraperitoneally injected with sodium nitrite to replicate AD model mice.At the same time,the corresponding solvent was poured into the stomach,except for distilled water in the normal control group and the model control group,the corresponding drug was poured into the stomach in other groups for 60 days.The subsequent water maze was used to detect the miee's escape latency and the number of crossing platforms.After the completion of water maze,brain tis- sue was taken from anesthetized mice,and the expression of TNF -α,AKT was detected by immunohistochemistry.Results Compared with the normal control group,the escape latency period of the model control group was significantly prolonged,the number of crossing the platform was significantly reduced,and the expressions of TNF -α,AKT were significantly increased.Compared with the model group,the above indexes were significantly improved in each dose group.Conclusion Gei Herba has an improved effect on d-galaetose combined with sodium nitrite replication in AD mice,which can down -regulate the expression of inflammatory factors and enhance the learning and memory ability of senile dementia mice.
