摘要
以水稻籼粳交F1 为材料 ,研究了改变培养基的氮素比例和碳源、30℃高温热击处理花药及改善供体植株的生理状况等措施对提高籼粳杂交后代花药培养力的影响。结果表明 ,在氮素比例中NH+4 NO-3 为5 .3 2 8.0时对提高花药培养力效果最好 ;混合使用 3%蔗糖和 3 %麦芽糖作碳源对提高出愈率、绿苗分化率及花药培养力效果显著 ;高温热击处理 36h ,可以同时使出愈率和绿苗分化率的提高达到显著水平 ;对供体植株的喷施 0 .8%KH2 PO4能使出愈率显著提高 。
Effects of some factors on the culture ability of rice anthers from indica/japonica hybridization were studied, which included changing carbon source and the ratio of nitrogen source, hot shocking the anthers at 30℃ and regulating physiologic state of donor rice plants. The results showed that the ratio of NH + 4/NO - 3 at to 5.3/28.0 was excellent in increasing anther culture ability; the carbon source with 3% maltose supplementing 3% sucrose could significantly increase frequency of callus induction, frequency of green plant differentiation and anther culture ability. When anthers were treated with hot shock at 30℃ for 36 h, frequency of callus induction and frequency of green plant differentiation were increased to significant difference level. At the same time, it was helpful for improving anther culture ability through increasing the frequency of callus induction to spray rice donor plants with 0.8% KH 2PO 4.
