结直肠癌进展中肿瘤浸润Th17细胞水平的变化 被引量:1

Changes of tumor infiltrating Th17 cells in the progression of colorectal cancer

摘要 目的探讨肿瘤浸润Th17细胞在结直肠癌进展中的变化及可能的机制。方法取28例结直肠癌患者的肿瘤组织,流式细胞术检测肿瘤浸润Th17细胞比例和在不同浓度的白细胞介素(IL)-1β、IL-6和转化生长因子(TGF)-β作用下体外培养的外周血单个核细胞(PBMCs)中Th17细胞比例;ELISA检测肿瘤组织培养液中IL-1β、IL-17A、IL-23、IL-6和TGF-β水平。结果与早期结直肠癌组织相比,晚期结直肠癌组织中Th17细胞比例及IL-1β、IL-17A、IL-23水平升高,但IL-6和TGF-β水平降低(P<0.05)。体外试验证实高浓度的IL-1β或高浓度的IL-6和高浓度的TGF-β能促进PBMCs中Th17细胞的增殖。结论 IL-1β、IL-6、TGF-β和IL-23可能调控结直肠癌进展中Th17细胞水平的变化。 [Objective] To explore the changes and possible mechanism of tumor infiltrating Thl7 cellsin the progression of colorectal cancer. [Methods] The tumor tissues were collected from 28 patients withcoloreetal cancer. The proportions of Th17 cells in tumor tissues and cultured peripheral blood mononuclearcells (PBMCs) treated by IL-1β, IL-6 and TGF-β in different concentrations were determined by flow cytometry; the levels of IL-1β, IL-17A, IL-23, IL-6 and TGF-β in supernatants from cultured tumor tissues weremeasured by ELISA. [Results] Compared with early tumor tissues, the proportions of Thl7 cells and the lev-els of IL-1β, IL-17A and IL-23 in advanced tumor tissues were significantly increased, but the levels of IL-6 and TGF-β were significantly decreased (P〈0.05). In vitro experiments confirmed that high concentration ofIL-1β or high concentrations of IL-6 and TGF-β could significantly increase the number of Thl7 cells inPBMCs. [Conclusion] The changes of Th17 cells in the progression of colorectal cancer are possibly modu-lated by IL-1β, IL-6, TGF-β and IL-23.
