摘要
目的复制大鼠心肌缺血再灌注模型,观察异丙酚联合贝那普利预处理对大鼠心肌缺血再灌注损伤的保护作用。方法采用结扎大鼠冠状动脉左前降支方法复制心肌缺血再灌注模型。大鼠随机分成5组:假手术组、缺血再灌注模型组、异丙酚预处理组、贝那普利预处理组及异丙酚和贝那普利联合预处理组。记录缺血再灌注前后不同时间点心率、血压及心律失常发生率;用流式细胞术检测心肌细胞凋亡率以及Bcl-2和Bax表达。结果异丙酚和贝那普利单独以及联合用药对心率、血压和血压-心率指数无明显影响。异丙酚和贝那普利联合用药后作用增强,可降低心律失常的发生率,减少心肌梗死面积,降低心肌细胞凋亡率,增高Bcl-2表达,降低Bax表达。结论异丙酚和贝那普利对大鼠心肌缺血再灌注损伤有协同的保护作用。
[Objective] To investigate the cardio-protective effect of the combination of Propofol and Be- nazepril preconditioning against myocardial ischemia reperfusion injury in rats. [Methods] All SD rats were randomly divided into five groups: sham group; myocardial ischmia/reperfusion group; Propofol group; Benazepril group; combination of Propofol and Benazepril group. These indexs were detected: heart rate (HR) and mean arterial pressure (MAP) were recorded. The infarct size was detected. Flow cytometry (FCM) was used to observe the apoptosis rate and the expression of Bcl-2 and Bax in cardiac myocyte. [Results] Propofol and Benazepril had no effect on HR, MAP and pressure-rate index. Propofol and Benazepril in combination decreased the incidence of arrhythmia, reduced myocardial infarct size, decreased the apoptosis rate of myocardial cells and the expression of Bax,and increased the expression of Bcl-2. [ Conclusion] Propofol and Benazepril have synergetic effect on proctecting cardiomyocyte from ischemia/reperfusion injury.
