摘要
目的采用制备高效液相色谱法,从蒙药大栀子中分离纯化其主要化学成分环烯醚萜苷类和西红花苷类,建立其纯化工艺。方法大栀子醇提物采用柱色谱分离,然后采用制备色谱系统,收集对应峰的流出液,并对产品进行了纯度检测。经理化常数测定,结合现代波谱学技术确定其结构。结果经过分离鉴定,得到两大类化合物,即环烯醚萜苷类与西红花苷类,6个单体化合物:栀子苷、去乙酰车叶草苷酸甲酯、鸡矢藤次苷甲酯、京尼平-1-β-龙胆苷,西红花苷-1、西红花苷-2。除栀子苷外,其它5种化合物均为首次从大栀子中分离得到。结论利用制备色谱技术进行蒙药大栀子化学成分的分离和纯化,具有简便、快捷、周期短、自动化程度高、收集全面等优点。
Objece Using preparative high performance liquid chromatography , and its main chemical components isolated and purified from the Mongolian medicine large gardenia are iridoid Glycosides and safflower glycosides , and establish its purifi- cation process. Methods Gardenia alcohol extract has been using column chromatography, and then using preparative chromatog- raphy systems , and collecting the effluent of the corresponding peak , and testing the purity of product. By testing the constant measurement , modern spectroscopic techniques combined with to determine its structure. Results After the isolation and identifi- cation of the two categories of compounds, namely iridoid glycosides and safflower glycosides , there were six monomer compounds , which were geniposide , deacetyl asperulisidic acid methyl ester , scandoside methyl ester, genipin - 1 - [3 - gentiobioside , cro- cinl , crocin - 2 . In addition to Geniposide , five kinds of other compounds are for the first time from large Gardenia in separa- tion. Conclusion Using preparative chromatography for the separation and purification of Mongolian medicine the large gardenia chemical composition , has a simple , fast , short cycle , high degree of automation , to collect comprehensive advantages.
