摘要
目的观察不同剂量硫氢化钠(NaHS)的抗动脉粥样硬化(AS)效应,综合评价外源性硫化氢(H2S)的作用。方法健康雄性SD大鼠50只,随机平均分为对照组、模型组、5.6 mg/kg NaHS组、2.8 mg/kg NaHS组和1.4 mg/kg NaHS组。采用高脂饲料结合给予维生素D3(VitD3)的方法制备AS模型。喂养12周后比较各组动物体质量、血脂、血H2S水平和主动脉病理变化。结果 5.6 mg/kg NaHS组体质量较模型组明显下降(P<0.05),且有4只动物死亡。模型组与对照组比较血清总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)明显升高(P<0.01),高密度脂蛋白胆固醇(HDL-C)升高(P<0.05);5.6 mg/kg和1.4 mg/kg NaHS组血脂水平与模型组比较无变化,2.8 mg/kg NaHS组TC和LDL-C降低(P<0.05),但仍高于对照组(P<0.01)。模型组H2S水平较对照组明显下降(P<0.01);3个NaHS组血清H2S水平均显著高于模型组(P<0.01),5.6 mg/kg NaHS组和2.8 mg/kg NaHS组H2S水平与对照组比较明显增高(P<0.01,0.05),1.4 mg/kg NaHS组与对照组比较无差异。模型组、5.6 mg/kg和1.4 mg/kg NaHS组主动脉均可见AS斑块,2.8 mg/kg NaHS组内皮略有损伤,内膜增厚,中层平滑肌细胞轻度增生,未见AS斑块。结论适当补充外源性H2S可提高AS动物血清H2S水平,降低血脂,抑制AS斑块形成。
[ Objective] To evaluate the effect of exogenous Hydrogen Sulfide (H:,S) on Atheroselerosis (AS) rats through different doses of sodium hydrosulfide (NariS). [ Methods ] Fifty healthy male SD rats were randomly divid- ed into five groups: control, AS model, 5.6 mg/kg NariS, 2.8 mg/kg NariS and 1.4 mg/kg NariS. A rat model of atherosclerosis was established by high fat diet and Vitamin D3. The body weight, blood lipid, blood H2S and aortic pathological changes were compared afterl2 weeks. [Results] The body weight of 5.6 mg/kg Naris group was sig- nificantly decreased than model, and there were four animals died in this group. The level of total Cholesterol (TC), low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C) in model group were increased significantly (P 〈0.01, P 〈0.05) compared with control group. Compared with model g'roup, lipid levels of 5.6 mg/kg group and 1.4 mg/kg group were no changes. The TC and LDL-C in 2.8 mg/kg group were decreased (P 〈 0.05) compared with model group, but still significant higher than control group (P 〈0.01). The level of H:,S in three Naris groups were higher than model group (P 〈0.01, P 〈0.05), which was decreased significantly compared with control group (P 〈0.01). The aortic AS plaque can be seen in model group, 5.6 mg/kg NariS group and 1.4 mg/kg Naris group. The 2.8 mg/kg blaHS group had significant improvement than model group with no AS plaque forma-tion. [ Conclusions ] Proper supplementation of exogenous H2S can increase the serum level of H2S, decrease the level of blood lipids and inhibit the formation of plaque in AS model animal.
