摘要
目的:研究人参总皂苷对脾虚模型大鼠的保护作用。方法:连续12周迫使大鼠在冷水([10±1)℃]中游泳至"力竭"并联合单日禁食双日高脂饮食以复制大鼠脾虚模型。实验分为正常对照(等容生理盐水)组、模型(等容生理盐水)组、人参总皂苷(50μg/kg)组,腹腔注射给药,每天1次,连续12周。检测大鼠体质量、体温,胸腺指数、脾脏指数与血清中白细胞介素(IL)-2、IL-6、肿瘤坏死因子α(TNF-α)的水平。结果:与正常对照组比较,模型组大鼠体质量、胸腺指数、脾脏指数显著降低(P<0.01或P<0.05),血清中IL-2、IL-6、TNF-α含量显著降低(P<0.01或P<0.05);与模型组比较,人参总皂苷组大鼠体质量、胸腺指数、脾脏指数显著升高(P<0.01或P<0.05),血清中IL-2、IL-6、TNF-α含量显著升高(P<0.01或P<0.05)。结论:人参总皂苷对脾虚模型大鼠有一定保护作用,其机制可能是通过提高细胞因子水平,恢复机体免疫功能,从而达到健脾益气的功效。
OBJECTIVE： To study protective effects of total saponins of Panax ginseng（TSPG） on spleen deficiency model rats. METHODS： Rats swam in cold water[（10 ± 1）℃] to be exhaustive for 12 consecutive weeks and were given fasting on odd days and high fat diet on even days to induce spleen deficiency model rats. Model rats were divided into normal control group （con- stant volume of normal saline）, model group （constant volume of normal saline） and TSPG group （50 μg/kg）. They were given in- traperitoneal injection of TSPG once a day continuous 12 weeks. Body weigth, body temperature, thymus index, spleen index, the levels of IL-2, IL-6 and TNF-α were detected. RESULTS： Compared with normal control group, body weigth, body temperature, thymus index and spleen index of model group were decreased significantly （P〈0.01 or P〈0.05） ; the levels of IL-2, IL-6 and TNF-α in serum were decreased significantly（P〈0.01 or P〈0,05）, Compared with model group, body weight, body temperature, thymus index and spleen index of TSPG group were increased significantly（P〈0.01 or P〈0.05） ; the serum levels of IL-2, IL-6 and TNF-α were increased significantly（P〈0.01 or P〈0.05）. CONCLUSIONS： TSPG can protect against spleen deficiency syn- drome. The mechanisms of TSPG treatment may be related to the improvement of cytokine and immune function so as to strengthen the spleen and replenish qi.
