摘要 目的:探讨TWEAK/Fn14在骨关节炎患者中的表达及其临床意义,为临床早期诊断骨关节炎探索新的方法。方法:收集35例骨关节炎患者为实验组,选择12例骨折患者作为对照组,采用ELISA方法检测两组人群血浆中TWEAK、RAN-TES、IL-18和IL-21水平,RT-PCR检测TWEAK mRNA在外周血单个核细胞和软骨细胞中的表达以及Fn14 mRNA在软骨细胞中的表达。结果:TWEAK、RANTES、IL-18、IL-21和ESR在实验组患者血浆中较对照组高表达(P<0.05)。在PBMC和软骨细胞中表达TWEAK mRNA,以及在软骨细胞表达Fn14mRNA,实验组均高于对照组(P<0.05)。结论:TWEAK在骨关节炎患者的血浆、PBMC和软骨细胞中高表达,Fn14在骨关节炎患者软骨细胞中高表达,表明TWEAK/Fn14可能参与了骨关节炎的病理发病过程,有望成为该病诊断的标志物。 Objective:To study the expressions and clinical significance of TWEAK/Fnl4 in patients with osteoarthritis and explore the novel diagnostic method for early osteoarthritis. Methods: 35 patients with osteoarthritis were collected as exprimental group and 12 fracture patients as control group. TWEAK, RANTES, IL-18 and IL-21expression in serum were detected by ELISA. TWEAK mRNA level was examined in peripheral blood mononuclear cells(PBMC) and chondrocytes by RT-PCR. Fnl4 rnRNA level was also examined in chondrocytes by RT-PCR. Results: TWEAK, RANTES, IL-18 and IL-21 expression were significantly higher in exprimental group than the normal group (P 〈 0.05). TWEAK mRNA expression in PBMC and chondrocytes, and Fnl4 mRNA expression in chondro- cytes were higher in exprimental group than the control group (P 〈 0.05 ). Conclusion: The increased expression of TWEAK and Fnl4 in patients with osteoarthritis suggests that TWEAK/Fn14 plays an important role in the pathogenesis of osteoarthritis. Furthermore, TWEAK and Fnl4 may be as a promising biomarker for diagnosis of the osteoarthritis.
