摘要
目的:研究天花粉蛋白对小鼠黑色素移植瘤血管生成拟态、PI3K mRNA及蛋白表达的影响。方法:小鼠接种B16细胞复制模型,接种结束后小鼠随机分为天花粉蛋白高、中、低剂量组(0.6、0.4、0.2 mg/kg)和空白对照组接受治疗。观察肿瘤一般状况,CD31与过碘酸-雪夫双重染色法检测血管生成拟态形态学,RT-PCR检测PI3K mRNA的表达,免疫组织化学检测PI3K蛋白的表达。结果:与空白对照组相比,天花粉蛋白高剂量组(0.6 mg/kg)瘤组织体积与重量、VM密度、PI3K mRNA与蛋白的表达均明显降低。结论:天花粉蛋白能够抑制小鼠黑色素移植瘤的血管生成拟态,推测可能与调控PI3K的表达有关。
Object:To investigate the effects of trichosanthin on the vasculogenic mimicry and the expression of PI3K in the melanoma transplant tumour. Method: The mouse melanoma transplant tumor model was made with B16 melanoma cell suspending liquid. After vaccination the mice were treated with trichosanthin at 0. 6 mg/kg,0. 4 mg/kg,0. 2 mg/kg and normal saline. The weight and volume of tumor was observed, vasculogenic mimicry was detected by CD31-PAS double stain,the expression of PI3K was detected by RT-PCR and immuno-histochemistry. Results: Compared with the control group,the weight and volume of tumor,vasculogenic mimicry density,expression of PI3K in high dose group(0.6 mg/kg) was obviously decreased(P 0. 05) . Conclusion: Trichosanthin could inhibit vasculogenic mimicry in the melanoma transplant tumour,which we supposed may be related with the regulation to expression of PI3K.
