摘要
目的探讨乳腺癌中是否存在血管生成拟态(VM),并通过研究其与基质金属蛋白酶-2(MMP-2)表达的相关性,初步探讨VM形成的机制。方法选取女性乳腺癌的石蜡标本共146例作为试验组,再选取20例女性乳腺的良性病变作为对照组,进行过碘酸雪夫氏反应以及CD34的双重染色,观察VM是否存在,再对VM阳性者和阴性者均进行MMP-2染色,比较二者相互关系。结果 VM的阳性表达与肿瘤大小、病理分级和淋巴结转移有关(P<0.05)。高分化和中低分化乳腺癌组织中VM的阳性率比较,差异有统计学意义(P<0.05)。在有淋巴结转移的乳腺癌组织中及无淋巴结转移的乳腺癌组织中VM的阳性率比较,差异有统计学意义(P<0.05)。MMP-2在乳腺癌组织中表达阳性率与乳腺良性病变比较,差异有统计学意义(P<0.05)。MMP-2在乳腺癌组织中的阳性表达率与病理学分级、淋巴结转移有关。高分化的乳腺癌组织MMP-2阳性率和中低分化乳腺癌组织比较,差异有统计学意义(P<0.05)。有淋巴结转移的乳腺癌组织MMP-2阳性率和无淋巴结转移的乳腺癌组织比较,差异有统计学意义(P<0.05)。MMP-2在VM阳性组中阳性表达率显著高于阴性组,二者在乳腺癌组织中的表达呈正相关性(r=0.550,P<0.05)。结论乳腺癌是中存在VM的,MMP-2表达同VM具有相关性。
Objective To investigate whether there exists vascularization mimesis（VM） in breast cancer,and to research the correlation between vascularization mimesis and expression of matrix metalloproteinase-2（MMP-2） in order to explore formative mechanism of VM.Methods One hundred and forty-six paraffin-embedded specimens of breast cancer were selected as trial group,and 20 benign breast lesion specimens were served as control group,All the specimens were stained by PAS and CD34,then VM was observed,moreover,these specimens were stained by MMP-2,and the correlation between positive VM and negative VM was analyzed.Results The positive expression of VM was correlated to tumor size,pathological grading,lymph node metastasis.There were significant differences in the positive expression rates of VM between well differentiated tissues of breast cancer and medium differentiated,poor differentiated tissues of breast cancer（P〈0.05）.There was a significant difference in the positive rate of VM between breast cancer tissue with lymph node metastasis and breast cancer tissue without lymph node metastasis（P〈0.05）.There was a significant difference in the positive expression rate of MMP-2 between breast cancer tissue and breast benign lesion（P〈0.05）.There were significant differences in the positiveexpression rates of MMP-2 among different pathological grades as well as with or without lymph node metastasis（P〈0.05）.There was a significant difference in the positive expression rate of MMP-2 between well differentiated tissues of breast cancer and medium differentiated,poor differentiated tissues of breast cancer（P〈0.05）.The positice expression rates of MMP-2 in positive VM group were significantly higher than those in negative VM group.Conclusion VM exists in breast cancer,moreover,the expression of MMP-2 is positively correlated to VM.
