摘要
目的采用乙醇注入-硫酸铵梯度法制备伊立替康长循环脂质体,并初步考察其体外抗肿瘤的生物活性。方法乙醇注入-硫酸铵梯度法制备伊立替康长循环脂质体,以结直肠癌细胞SW480,肝癌细胞HepG2,乳腺癌细胞MCF-7三种细胞为模型,MTT法检测伊立替康长循环脂质体对各种细胞的杀伤能力,同时采用荧光染色法,考察伊立替康长循环脂质体对乳腺癌细胞MCF-7形态及凋亡的影响。结果与游离伊立替康溶液相比,伊立替康长循环脂质体对人直肠癌细胞SW480,肝癌细胞HepG2,乳腺癌细胞MCF-7有明显的抗肿瘤活性,能够改变MCF-7细胞的形态,促进其凋亡。结论伊立替康长循环脂质体体外实验表明其能提高药物的抗肿瘤活性,其效果优于伊立替康注射液,体内实验有待于下一步进行。
【Objective】 Alcohol injection and ammonium sulfate gradient method was used to manufacture irinotecan loaded long circulation liposome,and investigate its biological activities.【Methods】 The human rectal cancer cell SW480,lung cancer cell HepG2,and breast cancer cell MCF-7 were used as model cells to examine the influences of different concentrations of CPT-11,CPT-11-loaded liposome,and blank liposome on growth inhibition.At the same time,the effects of CPT-11,CPT-11-loaded liposome,and blank liposome on the cell morphology of MCF-7 were investigated.【Results】 Both the CPT-11 and CPT-11 loaded long circulation liposome killed SW480,HepG2,MCF-7 in a concentration and time dependent manner in vitro.While at the same concentration,drug-loaded liposome had more potent killing function than that of free CPT-11.According to fluorescent microscopy,CPT-11 loaded liposome induced MCF-7 cell changed their morphology.【Conclusion】 Irinotecan loaded long circulation liposome could protect the lactone,prolonging the circulation time,and enhanced the anti-cancer activity compared to irinotecan injection.The in vivo study should be further investigated.
