摘要
为了探索百合幼胚离体培养的最佳胚性生长条件及植株再生途径 ,以授粉后 35 d的百合幼胚为外植体 ,进行了幼胚胚性生长 ,愈伤组织的诱导 ,愈伤组织不定芽、不定根的分化和再生植株的移栽试验 .结果表明 :在含有1 mg/ L IAA,1 m g/ L GA3,6 0 0 m g/ L CH的 MS培养基上培养 ,蔗糖含量为 6 %~ 8%时 ,百合幼胚胚性生长最好 ,培养 30 d后可以得到正常胚 .在含有 0 .1 m g/ L NAA,1 m g/ L BA的 MS培养基上可以形成淡绿色的愈伤组织 ,将这些愈伤组织转移到 1 / 2 MS无激素的培养基上培养可以形成大量的不定芽 ,不定芽生根移栽后 ,其成活率可达90
The inducing of embryonic growth and calli of immature embryo ,differentiation of calli and regeneration plant's transplantation were studied in 1999 by using 35 day old immature embryo of lily as the explant to provide a theoretic basis for the in vitro culture of immature embryo and regeneration of plant of lily .The results indicate that the 35 day old immature embryo of lily grows well on MS medium containing 1 mg/L IAA,1 mg/L GA 3,600 mg/L CH, 6%~8% sucrose.After cultured for 30 days, the immature embryo becomes normal mature embryo.Light green calli form the immature embryo cultured on MS medium containing 0.1 mg/L NAA, 1 mg/L BA. A lot of indefinite buds appear on the calli which are transferred to 1/2 MS medium without hormone. The survival rate of the regenerated plantlets which are transplanted is over 90%.
