基于ITS标记分析湖北省收集桑种质资源的亲缘关系 被引量:3

An Analysis on Genetic Relationship of Mulberry Germplasm Resources Collected in Hubei Province Based on ITS Marker

摘要 利用ITS标记从分子水平探讨湖北省境内收集的5个野生桑种、1个栽培桑种、1个变种共17份桑种质资源与外省区的6份桑种质资源之间的亲缘关系,为发掘和利用该地区特有的桑种质资源打下基础。结果表明,基于ITS标记的系统进化树与经典分类方法的结果接近,其中湖北省的4份华桑与四川省的川桑有较近的亲缘关系,同在一个分支;收集的3份蒙桑、1份变种鬼桑、1份鸡桑、5份白桑、1份山桑与广东桑、浙江省的瑞穗桑、云南省的滇桑之间有较近的亲缘关系,并与收集的2份长穗桑以及来自广西的钦州长果桑同在一个分支。利用DnaSP软件分析供试桑种质材料的ITS序列核苷酸多样性,符合被子植物ITS致同进化(concerted evolution)规律,变异主要发生在ITS1。利用AMOVA分析供试桑种质材料的遗传变异主要发生在种间,种间遗传变异率为79.49%,种内遗传变异率为20.51%(P<0.001)。分布于湖北省的桑种质资源处于抗寒屏障范围,是桑属植物迁移到此地后形成的类群。 Based on ITS sequence marker,the genetic relationship between 17 mulberry germplasm resources including 5 wild mulberry species,1 mulberry cultivar and 1 variant collected in Hubei province and 6 mulberry germplasm resources collected in other provinces was investigated at molecular level so as to provide references for exploration and utilization of unique mulberry germplasm resources in this region.The results show that the consequences of genealogical tree based on ITS sequence marker and of classical sorting method are close.4 Morus cathayana germplasms of Hubei Province have a close genetic relationship with Morus notabili of Sichuan Province,all of which formed a monophyletic clade.3 Morus mongolica,1 Morus mongolica var.diabolica,1 Morus australis,5 Morus alba and 1 Morus bombycis of Hubei Province have a close genetic relationship with Morus atropurpurea,Morus mizuho of Zhejiang and Morus yunnanensis of Yunnan.They clusted into one clade with 2 Morus wittiorm of Hubei Province and Morus laevigate of Qinzhou,Guangxi.DnaSP software was employed to analyze nucleotide diversity of ITS sequences of the tested mulberry germplasm resources,which was found to be correspondent with the concerted evolutionary pattern of angiosperm ITS.And nucleotide variation mainly happened in ITS1 sequence.AMOVA analysis revealed that variation of the tested mulberry germplasm resources mainly exists between species.The interspecific variation rate is 79.49% while the intraspecific genetic variation rate is 20.51%(P<0.001).Mulberry germplasm resources distributed in Hubei Province are within the protective range of cold resistance.They are the species formed after mulberry plants migrated into this area.
