微波消融对荷恶性黑色素瘤小鼠免疫功能的影响

摘要 目的:探讨微波消融对荷恶性黑色素瘤小鼠免疫状态的影响。方法:建立C57BL/6J小鼠B16黑色素瘤皮下移植模型,随机分为消融组和对照组。流式细胞术检测血清T淋巴细胞比例;免疫组化检测肿瘤组织CD68、CD49b的表达;ELISA法检测瘤内IFN-γ、IL-2、TNF-α含量;观察皮下移植瘤体积变化。结果:与对照组相比,消融组血清CD4+T淋巴细胞比例显著增高(P<0.01),CD25+T淋巴细胞比例显著降低(P<0.01),CD8+T淋巴细胞比例差异无显著性(P>0.05)。与对照组比较,消融组肿瘤组织CD68、CD49b的含量显著提高(P<0.01)。消融组IFN-γ、IL-2、TNF-α含量显著高于对照组(P<0.01)。在不同观察时间点,消融组皮下移植瘤体积均显著小于对照组(P<0.01)。结论:微波消融能上调抗肿瘤T淋巴细胞亚群,激活天然免疫细胞,增加抗肿瘤细胞因子,从而产生抗肿瘤效应。
