摘要
目的探讨兔肝VX2移植瘤治疗前后磁共振背景信号抑制弥散加权体部成像(diffusion weighted imagingwith background suppression,DWIBS)特征,对照分析肿瘤表观扩散系数(apparent diffusion coefficient,ADC)值与肿瘤细胞密度是否存在相关性。方法兔肝VX2移植瘤模型随机分为三组(8只/组)。建模后12天:TACE+ES组行TA-CE治疗和内皮抑素治疗;内皮抑素组注射内皮抑素治疗12天;对照组注射生理盐水12天。所有动物模型均于治疗前1天、治疗后第13天行DWIBS检查并记录各组肿瘤实质的ADC值。MR检查后取出肿瘤组织行HE染色并计算细胞密度。结果治疗后对照组ADC值低于TACE+ES组和内皮抑素组并有统计学意义(P=0.008,P=0.001);ADC值与肿瘤细胞密度间存在负相关性(r=-0.844,P=0.000)。结论 DWIBS结合ADC值的定量测量可以对活体肿瘤细胞密度进行无创性动态评价,有望为在体肿瘤分级提供新的帮助。
Objective To discuss the DWIBS appearance of the rabbit VX2 hepatic implantation tumor and analyze the relationship between apparent diffusion coefficient (ADC) value and cell density of tumor. Methods The rabbits bearing tumor were randomly divided into three groups, with eight rabbits in every group. 12 days after implantation, TACE and endostatin were used in TACE+ES group; only endostatin was administered via the ear vein in ES group; normal saline were administered via the ear vein in control group. The duration of the treatment all lasted 12 days. 1 day prior to as well as 13 days after treatment, DWIBS were performed. Apparent diffusion coefficient (ADC) values of the tumor were evaluated. Thirteen days after treatment, the rabbits were sacrificed immediately and the cell-density of the tumor was meas- ured according to HE. Results 13 days after treatment, the ADC values in control group was lower than those of TACE +ES group and endostatin group ( P d0.05). The density of the tumor cells had negative correlation with ADC value of the tumor( r =-0. 844, P = 0. 000). Conclusion DWIBS combining with ADC value can assess the cell density of tumor, which will be helpful for the tumor grade on living body.
