不同处理方法对菊花‘神马’脱除病毒的影响 被引量:7

Effects of Different Methods on Eliminating Cucumber Mosaic Virus(CMV) in Chrysanthemum morifolium ‘Shenma'

摘要 黄瓜花叶病毒是目前菊花普遍感染的病毒之一,为探讨其脱毒技术,以切花菊品种‘神马’为试验材料,采用茎尖处理(a)、病毒唑结合茎尖培养(b)、热处理结合茎尖培养(c)3种方法对其脱毒效果进行了研究。其中a处理分别剥取长度为0.2~0.3、0.4~0.5、0.6~0.8、0.9~1.0 mm的茎尖进行培养;b处理采用双因子完全随机区组的试验方法,对茎尖长度、病毒唑质量浓度进行筛选;c处理以昼夜变温并逐步升温的方式对试管苗进行处理。菊花母株及脱毒苗采用酶联免疫吸附测定法(ELISA)进行病毒检测。结果表明:a处理以0.4~0.5 mm长度的茎尖培养效果为宜;b处理采用茎尖长度为0.4~0.5 mm、病毒唑质量浓度为10 mg/L的组合最佳;c处理45 d后,剥取0.4~0.5 mm长度的茎尖培养最好。 Cucumber mosaic virus is one of the common viruses which infect Chrysanthemum. The experiment was conducted to discuss the virus elimination technique with the Chrysanthemum morifolium ' Shenma'. Shoot-tip treatment, shoot-tip with virazole treatment and shoot-tip with heat treatment were used to study the effect of virus elimination. The shoot-tips of 0.2-0.3 mm, 0.4-0.5 mm, 0.6-0.8 mm and 0.9-1.0 mm were cultured by the shoot-tip treatment. A randomized com- plete blocks design was conducted to screen the best shoot-tip length and virazole content by shoot-tip with virazole treat- ment. The test-tube plantlet was cultured by the heat treatment of different temperature step by step between day and night. ELISA test was used to identify the elimination efficiencies. The research effects showed that 0.4-0.5 mm shoot-tip culturing is the best in shoot-tip treatment, 0.4-0.5 mm shoot-tip culturing with 10 mg/L virazole is the best in shoot-tip with virazole treatment, and 0.4-0.5 mm shoot-tip after culturing 45 days heat treatment is the best in shoot-tip with heat treatment.
