摘要
目的:观察不同方法对吗啡成瘾后戒断大鼠戒断症状及NAc脑区多巴胺递质含量及其受体表达的具体影响机制。方法:采用逐日背部皮下注射盐酸吗啡注射液,末次注射后3 h给予纳络酮快速戒断,建立吗啡戒断SD大鼠模型。将50只大鼠随机分为对照组、模型组、电针组、拮抗组、针拮组,采取不同的方法治疗6天后,以ELISA法、原位杂交法分别检测NAc脑区DA含量及DA-D2RmRNA的表达。结果:与模型组比较,电针组能降低吗啡戒断后大鼠NAc脑区的DA含量(P<0.05);与模型组比较,电针组能调整戒断后大鼠NAc脑区DA-D2RmRNA的表达(P<0.05)。结论:电针可以改善吗啡戒断后症状,推测其在多巴胺系统通路中起到了类似于多巴胺受体激动剂的作用。
Objective:To observe the rats' withdrawal symptoms,the specific influence mechanism of dopamine content in NAc brain area and the expression of dopamine transmitter after morphine addiction. Methods:Establish a morphine dependence SD rat mode.We adopted hypodermic injection by morphine in back day by day,and injected naloxone in order to rapid withdrawal after the last injection three hours.ELISA and situ hybridization were used to observe the content of DA and the expression of DA-D2RmRNA in NAc brain area. Results:Electro-acupuncture could decrease SD rats' DA content in Nac obviously after morphine withdrawal(P0.05).Electro-acupuncture could adjust the expression of DA-D2RmRNA in NAc brain area after withdrawal(P0.05).Conclusion:Electro-acupuncture can ameliorate rats' symptoms after morphine withdrawal.We infer that Electro-acupuncture plays the role of dopamine transmitter excitomotor similarly in dopamine system.
