摘要
目的观察糖尿病大鼠肝纤维化发生情况,探讨内毒素和氧化应激水平在其中的作用。方法 60只Wistar大鼠随机分为正常对照组、糖尿病模型3个月组、6个月组和8个月组。一次性注射链脲佐菌素(STZ)诱导糖尿病模型。实验结束后处死大鼠,全自动生化分析仪检测肝功能、放射免疫法检测透明质酸(HA)、层黏蛋白(LN)、Ⅳ型胶原(4C)、Ⅲ型前胶原(PCⅢ),比色法检测内毒素(LPS)、游离脂肪酸(FFA)、丙二醛(MDA)、超氧化物歧化酶(SOD)、总抗氧力。肝组织HE、V-G染色观察病理改变。结果 3月组各检测指标与正常对照组比较没有显著变化,HE染色见肝细胞肿胀。6月组ALT、AST、LPS、MDA、SOD、总抗氧力与正常对照组;ALT、AST、MDA、SOD、总抗氧力与3月组比较差异有显著性。HA出现升高,细胞有脂肪变性。8月组除PCⅢ外各项指标与正常对照组、3月组比较差异有显著性,LPS、MDA、SOD、HA、LN与6月组比较差异有显著性,且内毒素水平与HA、4C显著相关,V-G染色出现纤维化。结论糖尿病有促肝纤维化发生趋势,内毒素和氧化应激代谢异常可能对其产生影响。
【Objective】To study the effect of endotoxin and oxidative stress on liver fibrosis in diabetic rats.【Methods】60 Wistar rats were randomly divided into control group,3 month group,6 month group,and 8 month group.Diabetes model were induced by one-shot injection of streptozotocin(STZ),and the rats were sacrificed at the end of the experiment.Liver function were detected by automatic biochemical analyzer,hyaluronic acid(HA),laminin(LN),Ⅳ collagen(4C),Ⅲ procollagen(PC Ⅲ) were measured by radioimmunoassay,and endotoxin(LPS),free fatty acid(FFA),malondialdehyde(MDA),superoxide dismutase(SOD),total antioxidant power were detected by chromatometry.Pathological changes of liver were observed by HE and VG staining.【Results】The indexes of 3 month group were no significantly changes compared with the control group,except liver cells swelling.It was significantly different from the control group in ALT,AST,LPS,MDA,SOD and total antioxidant power in 6 month group,and ALT,AST,MDA,SOD,total antioxidant power were significantly different compared with 3 month group,besides HA increased and cell degeneration.Compared with the control group and 3 month group,all the indicators excluding PC Ⅲ were significant difference in 8 month group.There was significant deviation between 8 month group and 6 month group for LPS,MDA,SOD,HA,LN,endotoxin levels were significantly associated with HA and 4C,and VG staining appeared fibrosis in 8 month group.【Conclusions】Diabetes can promote liver fibrosis,and it plays an important role in development of liver fibrosis and abnormal of endotoxin and disorder of oxidative stress could have effect on the liver fibrosis.
