摘要
目的糖尿病存在不同程度的氧化应激。文中通过复制1型糖尿病(type 1 diabetes mellitus,T1DM)大鼠模型,探讨氧化应激状态、血管内皮生长因子(vascular endothelial growth factor,VEGF)在糖尿病发生发展中的作用。方法 40只雄性SD大鼠随机分为T1DM组(23只)和对照组组(17只)。一定剂量的链脲佐菌素复制T1DM大鼠模型,采用比色法检测血清中的总抗氧化能力(total anti oxidation capacity,T-AOC)、超氧化物歧化酶(super-oxide dismutase,SOD)、谷胱甘肽(Glutathione,GSH)和丙二醛(Malondialdehyde,MDA);双抗体夹心ELISA法测定血清VEGF水平。结果 1成模7周后,T1DM组大鼠空腹血糖明显高于对照组大鼠[(23.01±3.62)mmol/L vs(5.08±0.46)mmol/L,P<0.05];血清中GSH、SOD水平明显低于对照组大鼠[1.88(0.36)g/L vs(3.82±1.05)g/L,(192.06±34.57)U/m L vs(242.37±32.79)U/m L,P<0.05];血清MDA、VEGF水平明显升高[(4.94±0.63)nmol/m L vs(4.42±0.37)nmol/m L,(23.97±1.84)pg/m L vs(22.32±1.71)pg/m L,P<0.05];组间T-AOC水平差异无统计学意义(P>0.05)。2实验大鼠空腹血糖与血清MDA、VEGF水平之间呈正相关(r=0.51,0.47),与血清GSH、SOD水平呈负相关(r=-0.71,-0.40);血清VEGF与MDA的水平呈正相关(r=0.32),与GSH水平呈负相关(r=-0.34)。结论 T1DM发生发展过程中,体内过氧化水平增高、抗氧化机制受损,这种变化与糖尿病高血糖状态密切相关;高水平的VEGF与糖尿病高血糖状态及过氧化程度增高有关。
Objective Different degrees of oxidative stress may exist in patients with diabetes mellitus( DM). The aim of this study was to explore the roles of oxidative stress and the level of vascular endothelial growth factor( VEGF) in the development and progression of DM in the rat model of type 1 DM( T1DM). Methods We established the T1 DM model in 23 male SD rats and included another 17 in the control group. We determined the total anti-oxidation capacity( T-AOC) and the levels of super-oxide dismutase( SOD),glutathione( GSH),and malondialdehyde( MDA) by chromatometry,and measured that of VEGF by ELISA.Results At 7 weeks after modeling,the level of fasting blood glucose( FBG) was significantly increased in the T1 DM rats as compared with the controls( [23. 01 ± 3. 62] vs [5. 08 ± 0. 46] mmol / L,P〈0. 05); the levels of GSH and SOD were markedly lower( 1. 88[0. 36]vs [3. 82 ± 1. 05]g/L and[192. 06 ± 34. 57]vs[242. 37 ± 32. 79]U/m L,both P〈0. 05) while those of serum MDA and VEGF remarkably higher in the T1 DM than in the control rats( [4. 94 ± 0. 63] vs [4. 42 ± 0. 37] nmol / m L and [23. 97 ± 1. 84]vs [22. 32 ± 1. 71] pg / m L,both P〈0. 05); no statistically significant differences were found in T-AOC between the two groups( P〈0. 05). FBG was correlated positively with MDA( r = 0. 51) and VEGF( r = 0. 47) but negatively with GSH( r =- 0. 71) and SOD( r =- 0. 40),while serum VEGF positively with MDA( r = 0. 32)and negatively with GSH( r =- 0. 34) in the experimental rats.Conclusion Peroxidation is increased and the anti-oxidation mechanism weakened during the development and progression of T1 DM,which is closely related to hyperglycemia. High VEGF is associated with hyperglycemia and enhanced peroxidation.
