藏药佐太中汞的长期蓄积性实验研究 被引量:15

摘要 目的探索KM小鼠以临床等效剂量长期给予佐太后汞在体内的蓄积情况,为佐太复方制剂临床用药安全提供科学依据。方法 KM小鼠分为空白组和给药组,给药剂量为每天6.67 mg.kg-1bw,给药4.5个月,停药1.5个月。分别于给药前、给药后0.5个月、1个月、1.5个月、2个月、2.5个月、3个月、3.5个月、4个月、4.5个月、停药0.5个月、停药1个月和停药1.5个月13个时间点采集血液、肾、脑、肝、脾,并检测汞含量。结果与空白组比较,给药组小鼠的肾脏中汞含量变化显著,在给药1.5个月后开始升高为38.24 ng/g,到3个月时达到最高值84.68 ng/g,停药1.5个月后逐渐恢复至正常水平31.46 ng/g;脑组织中汞含量在给药2个月后开始升高为23.60 ng/g,至4.5个月一直高于空白组,但数据呈波动状态,和空白组比较无统计学差异。血液、肝脏、脾脏中汞含量给药组较空白组变化不显著,停药0.5个月后汞含量接近空白组值。结论 KM小鼠长期给予临床等效剂量佐太后,汞在肾脏中产生一定蓄积,脑组织中的汞含量略高于空白组,血液、肝脏和脾脏中汞蓄积不明显,停药后各器官中的汞含量均可逐步恢复至正常水平。 Objective To explore mercury accumulation in KM mice following long-term use of Zuotai(gTso thal) and provide scientific basis for safety use of Zuotai in clinic.Methods KM mice were randomly divided into control groups and Zuotai groups,and the Zuotai groups were treated with Zuotai 6.67 mg·kg^-1 bwday for 4.5 months and drug discontinuance for 1.5 months.Blood,kidneys,brains,livers and spleens of mice were taken out every half month and the mercury content was measured.Results The accumulation of mercury in kidney in Zuotai groups was varied obviously compared with control groups,38.24 ng /g at 1.5 months and 84.68 ng / g at 3 months,the content of mercury become normal 31.46 ng / g when drug discontinuance for 1.5 months;the amount of mercury in brain was 23.60 ng / g at 2 months and was higher till 4.5 months,but the data were fluctuating.The mercury content in blood,liver and spleen didn’t vary strikingly compared with control groups and the amount of mercury decreased when drug discontinuance.Conclusion The mercury can be accumulated in kidney.The amount of mercury is higher in brain of Zuotai groups than control groups and won’t accumulate in blood,liver and spleen of KM mice following long-term treatment of clinical dose Zuotai,and the level of mercury can descent when drug discontinuance.
