摘要
目的:探索藏药佐太对慢性束缚应激抑郁模型小鼠的抗抑郁作用及其机制。方法:雄性小鼠分为空白组、模型组、佐太组(6.07mg/kg、60.7mg/kg、607mg/kg)、β-HgS组(3.38mg/kg),丙咪嗪(阳性药)组(15mg/kg),空白组和模型组每天灌胃2%淀粉悬浮液0.1ml/只,其它各组小鼠每天按照上述剂量灌胃0.1ml/只,同时进行慢性束缚应激造模。实验持续21天后,选用糖水偏爱实验和悬尾实验评价受试小鼠的抑郁样行为;通过观察小鼠海马区域组织形态,评价佐太对动物海马损伤的修复;利用ELISA法分析佐太对模型小鼠血清中的促炎性细胞因子白介素1β(IL-1β)、白介素6(IL-6)和肿瘤坏死因子(TNF-α)的影响。结果:与模型组比较,各佐太组(6.07mg/kg、60.7mg/kg、607mg/kg)、和β-HgS组(3.38mg/kg)小鼠悬尾实验中不动时间显著减少;海马组织病理切片发现,佐太(6.07mg/kg、60.7mg/kg、607mg/kg)和β-HgS(3.38mg/kg)可显著修复海马齿状回和CA1区的损伤;另外,发现各佐太组(6.07mg/kg、60.7mg/kg、607mg/kg)和β-HgS组(3.38mg/kg)小鼠血清中IL-1β,IL-6和TNF-α含量减少。结论:佐太可显著改善CRS模型小鼠的抑郁样行为,可能是通过降低小鼠脑内促炎性细胞因子IL-1β,IL-6和TNF-α的分泌而实现。
Objective: To study the antidepressant effect and mechanism of Zuotai. Method:70 male KM mice were randomly assigned into 7 groups as control(2% starch), model(2% starch), Zuotai (6.07,60.7,607mg/kg), β-mercuric sulfide(3.38mg/kg) and imipramine (15mg/kg). All mouse were exposed to chronic stress (CRS) for 21 consecutive days. Tail suspension test and sucrose preference test were applied to assess the antidepressant efficacy, hippocampus slides were observed and IL-1β, IL-6, TNF-α level concentrations in serum were measured by ELISA kit. Result: Zuotai 6.07 mg/kg, 60.7 mg/kg,607mg/kg can significantly short the motionless time in Tail Suspension Test( P 〈 0.05 ,P 〈 0,01 ) and can increased sucrose consumption index. Depression-like mouse's hippocampus was significantly improved by Zuotai 6.07 mg/kg,60.7 mg/kg,607mg/kg . In addition, Zuotai 6.07 mg/kg,60.7 mg/kg,607mg/kg can significantly lower IL-1β, IL- 6 and TNF-α level in serum in CRS mouse ( P 〈 0.01 ). Conclusion: Zuotai showed the antidepressant effect on CRS mouse and it may work by decreasing pro-inflammatory cytokines.
