接骨膏对家兔骨折愈合BMP-7表达的研究 被引量:16

The experimental study of Tujia Medicine Jiegu Paste(土家族药接骨膏) on rabbits fracture healing BMP-7 expression

摘要 目的:探讨土家族药接骨膏在家兔骨折愈合中对BMP-7表达的影响。方法:48只日本大耳白兔制作左侧桡骨3mm骨缺损模型,随机分为接骨膏实验组(简称实验组)和模型对照组(简称对照组),每组24只。实验组给予接骨膏外敷治疗,对照组不予治疗。于术后2W、4W、6W处死实验动物,每组动物处死前行DR摄片,然后取骨标本组织切片,分别进行HE染色和免疫组化检测以观察骨折愈合情况和BMP-7在治疗过程中的变化。结果:与对照组比较,实验组骨痂出现时间较早,数量多,骨痂阴影密度高,且骨性愈合时间和骨髓腔再通较早;实验组纤维连接和纤维骨痂形成较早,成骨细胞数量较多,骨痂及骨性愈合出现时间较对照组早,骨痂面积大,生长速度较快;免疫组化MOD值与对照组差异有显著性(P<0.01)。结果表明,以5.0g/kg(体重)接骨膏外敷能明显促进家兔骨折局部BMP-7表达和骨痂形成,有效的促进骨折愈合。结论:接骨膏通过促进骨折局部BMP-7的表达是促进骨折愈合的重要作用机制。 In this paper,we will investigate file expression effects to BMP-7 concerning with Tujia Medicine Jiegu Paste on rabbits of infracture healing. Methods: 48 rabbits are selected to make left radius 3mm bone defect model, and are randomly allocated into Jiegu paste experimental group (the experimental group) and model control group ( control group) ( each group 24 cases ). The experimental group is treated with Tujia Jiegu Paste for compresses, while the control group not treatment. After the operation, 2W ,4W, 6W experimental animals are to be put to death. Animals in each group are carried on DR-radiography before death, and then take the bone specimens tissue slices to conduct HE dyeing and immunohistochemieal examination for observing the fracture healing and changes on BMP-7 during the course of treat- ment, respectively. Results:( 1 ) DR-radiography:Compared with the control group,bone callus in the experimental group appear earlier, with more numbers and a higher shadow density, and bone healing and bone marrow cavity recanalization also happen earlier;(2) HE dyeing:Fi- brous connection and fiber bone callus in the experimer^tal group form earlier,with larger number of osteoblasts. Bone callus and bone healing occur earlier than the control group ,with the larger bone callus area and a faster growth speed; (3) Concerning twosample mean t-test compar- ison between Immunohistochemical Mean optical density (MOD) values and the control group row, there exists significant difference ( P 〈 0. 01 ). The results show that,5.0g/kg (weight) give Jiegu Paste for external application, Can obvious promote rabbit fracture local of BMP-7 ex- pression and callus formation, effective in promoting fracture healing. Conclusion: Tujia medicine Jiegu paste, according to promote fractures local BMP-7 expression,is the major mechanism to boost the fracture healing.
